微生物用于吐温80/磷酸钾体系的萃取生物转化

摘要 对生物转化所必须的目标菌种进行了筛选和培养 ,制备了棒状杆菌种液 .在吐温 80 /磷酸钾液 -固萃取体系中 ,用此菌种液 ,催化丙烯腈为丙烯酰胺 .确定了用气相色谱法 ,作为生物转化产物的检测手段 .结果表明 ,丙烯酰胺的色谱响应值在 ( 0 .5~ 3.0 )× 10 -3 μg呈线性 ;总盐浓度 1.89mol·L-1,K2 HPO4 :KH2 PO4 为 3∶ 2 ,Tween80 :10 % ,p H7.0 0 ,产物分配在吐温固相 ,底物、菌种留在富盐水相 .上述条件 ,有利于丙烯腈转化为丙烯酰胺 ,可用倾出法 ,方便快速移走产物 ,从而实现腈的高转化率 . The target bacterium for the extractive bioconversion were selected and cultured. A cotynebactrium solution was produced.In the Tween80 /potassium phosphate liquid- solid extraction system,the biocatalyst of acrylamide from acrylonitrile was carried out by using this bacterium.The bioconversion product is detected by gas chromatography.The linear range GC response for acrylamide is (0 .5~ 3.0 )× 1 0 - 3 μg.In the liquid- solid extraction system the total salt concentration is1 .89mol· L- 1 ;K2 HPO4:KH2 PO4=3∶ 2 .The Tween 80 concentration is 1 0 % .The system acidity is p H 7.0 .Under above conditions the product was selectpartied to Tween80 solid phase,substrate and bacterium were remained in the rich salt aqueous phase.The product is conveniently and rapid transferred with decantation.The above process and conditions increase the productvity of acrylamide from acrylonitrile.
