苦参素对食管癌Eca-109细胞BIP和CHOP mRNA表达的影响 被引量:11

摘要 目的:探讨苦参素对食管癌Eca-109细胞内质网应激相关基因免疫球蛋白重链结合蛋白(BIP)和C/EBP环磷酸腺苷反应元件结合转录因子同源蛋白(CHOP)表达的影响。方法:体外培养食管癌Eca-109细胞,分为空白对照组、5-氟尿嘧啶组(2 g/L)和不同剂量的苦参素组(2.0、1.0、0.5 g/L)。采用MTT法检测细胞增殖能力,Annexin V-PI法检测细胞凋亡率,实时荧光定量PCR法检测BIP和CHOP mRNA的相对表达量。结果:各组Eca-109细胞吸光度值、凋亡率、BIP和CHOP mRNA相对表达量比较,差异均有统计学意义(F=3.883、476.221、129.331、14.139,P均<0.05)。与空白对照组相比,苦参素组和5-氟尿嘧啶组Eca-109细胞吸光度值降低,凋亡率升高,BIP mRNA相对表达量降低,CHOP mRNA相对表达量升高(P均<0.05)。结论:苦参素可抑制Eca-109细胞增殖,促进其凋亡,BIP基因可能是其作用靶点之一,凋亡过程由CHOP介导。 Aim : To explore the effects of oxymatrine on the expression of immunoglobulin heavy chain binding protein (BIP) related endoplasmie retieulum stress (ERS) gene as well as CAAT/enhaneer binding protein homologous protein (CHOP) of esophageal carcinoma Eta-109 cells. Methods:The experiment was divided into control group, 5-fluorouracil group(2 g/L) and oxymatrine groups(2.0, 1.0, 0.5 g/L). Eca-109 cells were cultured while intervened with different concentrations of oxymatrine. Cell proliferation(A value) and apoptosis rate were detected by MTT and Annexin V-PI doub- le staining, then the expressions of BIP and CHOP mRNA were detected by real-time quantitative PCR. Results: There was significant difference in A value, apoptosis rate, the expressions of BIP and CHOP mRNA among the five groups ( F = 3. 883, 476. 221 , 129. 331 , 14. 139,P 〈 0.05). Compared with control group, the A value of the other four groups de- creased, apoptosis rate increased, the expression of BIP mRNA downregulated, and the expression of CHOP mRNA upreg- ulated (P 〈 0.05 ). Conclusion : Oxymatrine could inhibit proliferation and induce apoptosis of Eea-109 cells, and BIP gene may be one of the targets in the action, while CHOP mediates the apoptosis.
