摘要
肿瘤血管为肿瘤发生、发展提供足够的氧分和营养,靶向肿瘤血管新生可以达到"饿死"肿瘤的治疗效果。但是肿瘤血管新生的分子调控机制非常复杂,需要许多生长因子、受体及信号通路的介导。尽管已经有很多抗血管新生的靶向药物上市或正在被研发,然而有些药物的临床治疗效果却差强人意,有的还存在耐药问题,这可能就是肿瘤血管新生有多种调控途径导致。因此本文将通过综述肿瘤血管相关的分子调控机制,探讨血管生成环节中可能的抗肿瘤靶点,阐述目前靶向肿瘤血管新生药物的研究情况和存在的问题,并且提出肿瘤血管研究的新策略。
Tumor blood vessels provide sufficient oxygen and nutrients for tumor growth and development, so drugs targeting tumor angiogenesis can “starve” and inhibit tumor. However, the molecular regulation mecha- nism of tumor angiogenesis is very complicated, which may be mediated by some growth factors, receptors and sig- nal pathways. In addition, many antiangiogenic drugs have been approved or are being developed, but the clinical therapeutic effects of certain drugs are limited. Moreover, there exist drug resistance problems sometimes, which may be explained by complicated and confused regulation pathways of angiogenesis. Therefore, in this article we re- viewed the related molecular regulation mechanisms of tumor angiogenesis, explored possible antiangiogenic targets in angiogenic pathways, elaborated the development of drugs through inhibiting tumor angiogenesis and the existing problems, and put forward new strategies for researches of tumor angiogenesis.
