摘要
目的:建立以高效液相色谱法同时测定山楂丹参分散片中丹参素钠、原儿茶醛、金丝桃苷、迷迭香酸、紫草酸、丹酚酸B的含量测定方法。方法:采用Phenomsil-C18(250mm×4.6mm,5μm)色谱柱,流动相为乙腈-甲醇-0.5%磷酸水溶液,梯度洗脱,流速:1.0mL·min^-1,检测波长:0~33min为280nm,33~56min为257nm,56~85min为289nm,柱温:30℃。结果:在本色谱条件下,丹参素钠、原儿茶醛、金丝桃苷、迷迭香酸、紫草酸、丹酚酸B的进样量分别在0.05016~0.2508μg,0.02890~0.1446μg,0.01520~0.07620μg,0.1146~0.5732μg,0.2631~1.315μg,1.972~9.86μg范围内与色谱峰面积呈良好的线性关系;加样回收率(n=6)均在97.7%~102.8%,RSD均小于3.0%。结论:本方法快速、准确,重复性好,可以满足山楂丹参分散片的含量测定要求。
Objective: To establish an HPLC method for determination of sodium danshensu, protocate- chuie aldehyde, hyperoside, rosmarinic acid, lithospermic acid and salvianolic acid B in dispersible tablets of haw- thorn and salvia miltiorrhiza. Methods: The separation was performed on a Phenomsil-C18 ODS (250 mm× 4.6 mm, 5 μm) column with the gradient elution of acetonitrile-methanol-0.5% phosphoric acid water solution at the flow rate of 1.0 mL·min^-1 The detection wavelengths were 280 nm for sodium danshensu, protocatechuie aldehyde and hyperoside, 257 nm for rosmarinic acid and lithospermic acid, and 289 nm for salvianolic acid B. The column tem- perature was set at 30℃. Results: Sodium danshensu, protocatecbuic aldehyde, hyperoside, rosmarinic acid, lithospermic acid and salvianolic acid B had good linearity in the ranges of 0. 050 16 ~0. 250 8 μg, 0. 028 90 ~ 0. 144 6 μg, 0. 015 20~0.076 20 μg, 0. 1146~0. 5732 μg, 0. 2631 ~1. 315 μg and 1.972~9. 860 μg, respec- tively. The average recoveries of all analytes were 97.7% - 102.8% with RSDs less than 3.0%. The contents of sixmarker compounds in hawthorn and salvia miltiorrhiza were different. Conclusion: The developed method is rap- id and accurate with high repeatability and stability, which meets the content determination requirements of dispers- ible tablets of hawthorn and salvia miltiorrhiza.
