摘要
目的:观察六味地黄丸对2型糖尿病大鼠周围神经病变的治疗作用。方法:雄性SD大鼠高脂高糖饲料喂养8周后腹腔注射20 mg·kg-1链脲佐菌(STZ),制备2型糖尿病大鼠模型;将造模成功大鼠分成模型组、糖末宁对照组、六味地黄丸高、中、低剂量组。给药8周后检测大鼠坐骨神经的传导速度并对大鼠坐骨神经组织进行HE染色切片观察;测定坐骨神经组织醛糖还原酶(AR)和Na+-K+-ATP酶的活性。结果:与模型组相比六味地黄丸高剂量组大鼠坐骨神经传导速度明显提高(P<0.01);坐骨神经组织病理检查显示:模型组大鼠周围神经纤维分布稀疏,神经髓鞘细胞有变性肿胀和坏死缺失,有明显脱髓鞘现象,六味地黄丸高、中剂量组与模型组比较,纤维分布较密,脱髓鞘现象较轻。模型组大鼠坐骨神经组织AR活性明显增高、Na+-K+-ATP酶的活性降低。六味地黄丸高剂量组可降低AR活性,提高Na+-K+-ATP酶活性。结论:六味地黄丸对2型糖尿病大鼠周围神经病变有一定保护作用。
Objective: To determine the effect of LiuWeiDiHuang pill on peripheral neuropathy in rats with type 2 diabetes. Methods: Type 2 diabetes was induced by high-fat and high-sugar fed, and an intraperitoneal in- jection of STZ 20 mg· kg^- 1 8 weeks later. The rats meeting type 2 diabetic standards were selected, and treated with Tangmoning and 3 doses of LiuWeiDiHuang pill. At 8th week after the treatment, nerve conduction velocity (NCV) was measured, and the rat sciatic tissue structures were observed. The aldose reductase ( aldose reductase, AR) and Na ^+-K ^+-ATPase activities in the sciatic nerve tissue were determined. Results: Compared with diabetic control, the NCV was accelerated in rats with large dose of LiuWeiDiHuang pill (P 〈0.01 ). The nerve fibers were much less with nerve Schwann cell degenerating, swelling, necrosis and missing in diabetic rats. Large and mod- erate doses of LiuWeiDiHuang pill attenuated the changes in the nerve fibers. AR activity increased and Na ^+ -K ^+ ATPase activity reduced in diabetic rats. Large dose of LiuWeiDiHuang pill attenuated the changes in these en- zyme activities. Conclusion: LiuWeiDiHuang pill has a protective effect on peripheral neuropatby in rats withtype 2 diabetes.
