摘要
该研究采用绿色木霉AS3.30 32 (Trichodermaviride)液体发酵生产纤维素酶 ,研究了碳源、氮源、培养基起始 pH值、接种量、摇床转速对绿色木酶产酶活力的影响 .结果表明 :①以爆破后的甘蔗渣为碳源时滤纸酶活和 β Case酶活力分别高达 5 .37U/mL和 4.89U/mL ;②不同氮源产酶活力大小顺序为 :NH4 NO3,(NH4 ) 2 HPO4 ,(NH4 ) 2 SO4 ,尿素 ,NH4 Cl,酵母膏 ;③培养基起始 pH为 3 .5 ,摇床转速为 15 0r/min ,培养温度为 2 8℃时 ,产酶活力最高 ;④接种量对产酶活力影响不大 ,以体积分数 φ为 5
Cellulase production was carried out by submerged fermentation with Trichoderma viride AS?3 3032.The effects of nitrogen source, carbon source, initial pH of medium, inoculation and velocity of shaker on cellulase synthesis are studied. The results are:① using steam exploded bagasse as carbon source results in very high enzyme production in shake flask culture, 5 37?U/mL, β Glucosidase activity 4 89?U/mL, respectively; ② The order of the cellulase activity formed was: NH 4NO 3,(NH 4) 2HPO 4,(NH 4) 2SO 4,urea,NH 4Cl and yeast extract; ③ The optimum conditions of producing cellulase were: the initial pH was 3.5, the shaker was 150?r/min, the temperature was 28?℃; ④ The impact of amount of inoculation on cellulase production is little, the 5% inoculation ( V/V ) suitable.
