摘要
目的 :探讨中药复方福尔肝Ⅵ号对大鼠肝纤维化的作用。方法 :二甲基亚硝胺腹腔注射诱导大鼠肝纤维化模型 ,观察各组大鼠体重、肝脾重、腹水及门静脉压力等变化 ,HE染色及丽春红胶原染色观察肝组织炎症与胶原沉积病理改变 ,检测血清肝功能及肝组织羟脯氨酸含量。结果 :模型大鼠体重与肝重减轻 ,脾脏增大 ,腹水明显 ,血清ALT与AST水平、总胆红素含量升高 ,白蛋白下降 ,肝组织炎症坏死与胶原沉积明显 ,纤维间隔形成 ,Hyp含量增多。经福尔肝Ⅵ号及γ -干扰素治疗后 ,模型大鼠体重与肝重趋于恢复 ,腹水与门静脉压力减轻 ,血清AST水平与总胆红素含量显著下降 ,肝组织胶原沉积与Hyp含量明显减少。两药物作用无明显差别。结论 :福尔肝Ⅵ号具有较好抗DMN模型大鼠肝损伤及肝纤维化的作用 ,抗炎与抑制胶原过度增生沉积可能是该方抗肝纤维化的主要作用机理目的 :探讨中药复方福尔肝Ⅵ号对大鼠肝纤维化的作用。方法 :二甲基亚硝胺腹腔注射诱导大鼠肝纤维化模型 ,观察各组大鼠体重、肝脾重、腹水及门静脉压力等变化 ,HE染色及丽春红胶原染色观察肝组织炎症与胶原沉积病理改变 ,检测血清肝功能及肝组织羟脯氨酸含量。结果 :模型大鼠体重与肝重减轻 ,脾脏增大 ,腹水明显 ,血清ALT与AST水平、总胆红素含量升高 ,白蛋白下?
Objective:To investigate the effects of a Chinese herb formula,Fugan Ⅵ on rat's liver fibrosis induced by dimethylnitrosamine. Methods:The fibrotic model was induced by ip dimethylnitrosamine (DMN) in ra ts,then administrated with Fugan Ⅵ qd for 4w and treated with interferon- γ(IFN-γ) as control drug.The weights of body,liver and spleen,ascites and port al vein pressure was observed.the liver inflammation and collagen deposition wer e observed with HE and ponceau stain.Serum liver function test,hepatic hydroxypr oline (Hyp) was measured. Results:In fibrotic rats,the weights of body and liver decreased,spleen weight a nd portal vein pressure increased,and ascites formed.Serum ALT and AST activitie s and total bilirium content increased,but albumin level decreased.The liver inf lammative necrosis and collagen deposition were obvious,some fibrotic septa form ed and Hyp increased.Both Fugan Ⅵ and γ-IFN restored body and liver weigh t,reduced ascites and portal vein pressure,decreased the serum T.bil content and AST level,decreased the hepatic collagen deposition and Hyp content significant ly.Two drugs had no obvious difference on the actions. Conclusions:Fugan Ⅵ recipe has effects against liver injury and fibrosis i nduced by DMN in rats,the mechanisms of the recipe action against fibrosis were associated with its inhibition of liver inflammation and excessive collagen prod uciton.
