摘要
大曲药作为白酒生产重要的菌酶混合制剂,为后续的窖池发酵提供了主要的酶制剂来源。针对浓香型春季大曲,以液化酶酶活力作为参考指标,研究了水溶液浸提的pH、温度、料液比、提取时间等条件,经正交实验并验证,得到了提取的最佳条件为pH6.0、温度30℃、料液比1:2、提取时间2 h。对提取物料处理方式进行了研究,对浸提液进行超声波破碎处理有利于产孢子微生物分泌的纤维素酶的释放。对于液化酶、糖化酶、中性蛋白酶等主要是细菌来源酶类,物料、浸提液进行超声波破碎处理,均没有显著效果。
Daqu as liquor production important materials of bacteria and enzyme, provides a major source of enzyme for brewing. This study aimed at spring Luzhou-flavor Daqu, liquefied enzyme was used as a reference, the pH, temperature, liquid ratio, extraction time of aqueous extraction were studied, through the orthogonal experiment and verification, the best extraction conditions was pH 6.0, 30 ℃, solid-liquid ratio 1:2, extracting time 2 h. The extraction of materials handling was studied, the higher cellulose enzyme activity was obtained in leaching solution by ultrasonic crushing processing, this cellulose enzyme was considered to derived from the spore production microorganisms. The α-amylase, glucoamylase, dispase were considered to derived mainly from bacteria, to material, ultrasonic crushing leaching solution, and have no significant effect.
