摘要
对库尔勒香梨中脂氧合酶活性测定条件进行了研究,结果表明:粗酶液中添加1.5%PVP,35℃条件下,亚油酸溶液40μL,加入pH7.0磷酸二氢钠-磷酸氢二钠缓冲液,于234 nm波长条件下检测脂氧合酶活性最为理想;在确定酶活条件的基础上,测定其在冷藏、涂膜贮藏和气调贮藏期间库尔勒香梨中的脂氧合酶活性的变化规律,结果表明在气调贮藏条件下脂氧合酶活性相对较低,对脂氧合酶的活性具有较为显著的抑制作用。
In this experiment, determination of lipoxygenase(LOX) activity of Korla pear was studied. The optimal determination conditions were as follows: adding 1.5% PVP and NaH 2 PO 4-Na 2 HPO 4 buffer(pH7.0) into crude enzyme solution, when its temperature reached 35 ℃, adding 40 μL LA solution, then analyzing the enzyme activity at 234 nm wavelength, under these conditions LOX activity is ideal. According to the optimal conditions, the changing rules of LOX activity of the Korla pear were studied during the period of chilling temperature storage, painting film storage and controlled atmosphere storage, the inhibiting effect of controlled atmosphere storage was more evident.
