益气逐瘀利水方对破裂型腰椎间盘突出退变髓核细胞Col Ⅱ及Aggrecan mRNA的影响

The effect of Yiqi Zhuyu Lishui Formula on Col Ⅱ and Aggrecan mRNA in Degenerative Nucleus Pulposus Cell of Ruptured Lumbar Disc Herniation

目的:探讨益气逐瘀利水方对人体破裂型腰椎间盘突出退变髓核细胞中Ⅱ型胶原(ColⅡ)、聚集蛋白聚糖(Aggrecan)mRNA的表达的影响。方法:选择破裂型LDH的住院患者经手术摘除的腰椎间盘组织,采用组织块法培养原代细胞;以SPF级雄性SD大鼠制备药物血清。将传代第1代的退变髓核细胞分为3组,实验组分别予体积比为5%、10%的益气逐瘀利水方血清DMEM培养液;对照组加入生理盐水血清DMEM培养液。通过Realtime PCR检测ColⅡ、Aggrecan mRNA的表达。结果:与生理盐水组比较,体积比为10%的益气逐瘀利水方血清可上调破裂型腰椎间盘突出退变髓核细胞中ColⅡ、Aggrecan mRNA的表达。(P<0.05)结论:益气逐瘀利水方可能通过影响破裂型腰椎间盘突出髓核细胞中ColⅡ、Aggrecan含量,进而发挥防治破裂型腰椎间盘突出髓核细胞的退变作用。

Objective：To discuss the effect of Yiqi Zhuyu Lishui Formula on Col Ⅱ and Aggrecan mRNA in degenerative nu- cleus pulposus cell of ruptured lumbar disc herniation （LDH）. Methods： Lumbar intervertebral disc tissues were obtained from hospitalized patients with ruptured LDH who underwent surgery. Primary cells of degenerative nucleus pulposus were cultured using method of tissue piece. Drug serum was prepared from SPF male SD rats. The first-generation degenerative nucleus pulposus cells were divided into three groups, which were respectively cultivated in Yiqi Zhuyu Lishui Formula DMEM serum with volume ratio of 5 %, 10% arid physiological saline. The Col Ⅱ and Aggrecan mRNA were detected by Realtime PCR. Results：Compared with the physiological saline group and 5% Yiqi Zhuyu Lishui Formula DMEM serum group, the group of 10% promoted the expression of Col Ⅱ and Aggrecan mRNA in degenerative nucleus pulposus cells （P 〈0.05）. Conclusion： Yiqi Zhuyu Lishui Formula may prevent the degeneration of nucleus pulposus cell via affecting the ex- pression of Col Ⅱ and Aggrecan in ruptured lumbar intervertebral disc.
