云南省新型肠道病毒EV80的基因特征分析 被引量:1

摘要 目的对云南省3株新型肠道病毒EV80的基因特征进行分析。方法对3株EV80病毒VP1区基因进行RT-PCR和基因测序,并在NCBI、EBI和DDBJ三大基因库搜索EV80VP1区基因序列,用MEGA5.0软件计算所得序列的核苷酸和氨基酸相似性,并做基因树进行基因特征分析。结果共检索到35株EV80的VP1区基因序列,去除序列太短不能分析的2株,对云南株和基因库中共33株进行了分析。33株病毒可分为4个基因型,原型株USA-CA67-10387单独聚成一组,为基因型A,云南株和山东株同属基因型D,为中国特有的基因型。结论虽然目前全世界新型肠道病毒EV80的分离数不多,但其地理分布较为广泛,不同国家、地区的毒株大都为不同的基因型,具有基因多样性特点。本研究对今后中国开展新型肠道病毒EV80的分子流行病学研究和传播链的发现和疫情处置提供了实验室基础。 Objective To analyze the genetic characteristics of the newer enterovirus EV80 isolated from Yunnan province of China. Methods The VP1 gene of three EV80 Yunnan strains was sequenced and compared with other available sequences from NCBI, EBI and DDBJ; the nucleotide and amino acid identities and genetic characteristics were analyzed. Results There are altogether 35 strains sequences were obtained and of which, 2 strains were excluded because their sequences were too short. Yunnan strains and other 30 strains were analyzed. The 33 EV80 strains can be divided into 4 genotypes, the prototype strain"USA-CA67-10387" was solely clustered into genotype A; Yunnan and Shandong strains were grouped into genotype D. Conclusions Currently, there are only a few EV80 strains were isolated that are widely distributed around the world. Strains isolated from different countries were classified into different genotypes, demonstrating multiple genetic characteristics.
