MMP-2和CXCR4mRNA在人肾透明细胞癌中的表达及意义 被引量:1

摘要 目的研究人肾透明细胞癌(ccRCC)组织中基质金属蛋白酶-2(MMP-2)和CXC趋化因子受体4(CXCR4)的表达情况及相关性,探讨二者与临床病理因素的关系。方法应用反转录-聚合酶链式反应(RT-PCR)方法检测68例ccRCC组织和36例癌旁正常肾组织中MMP-2与CXCR4mRNA的表达水平。结果在ccRCC组织中MMP-2和CXCR4mRNA的半定量值分别为0.848±0.367和0.939±0.336,高于在正常肾组织中的半定量值0.379±0.138和0.418±0.146,差异有统计学意义(t=7.375、8.860,P<0.05);在ccRCC中MMP-2和CXCR4mRNA的表达与肿瘤的临床分期、分化程度和淋巴结转移情况有关(t=2.169~3.852,P<0.05),而与患者的年龄、性别及肿瘤大小无关(t=0.201~0.630,P>0.05);在ccRCC组织中MMP-2和CXCR4mRNA的表达呈正相关(r=0.832,P<0.05)。结论 MMP-2和CXCR4可能共同参与了人ccRCC的发生、发展、侵袭和转移,并且在发挥作用时具有协同作用,联合检测MMP-2和CXCR4可能有助于协助ccRCC的早期诊断和病情评估。 Objective To investigate the expressions and correlation of matrix metalloproteinases-2 (MMP-2) and CXC chemokine receptor 4 (CXCR4) mRNA in human clear cell renal cell carcinoma (ceRCC) tissues,and to study their association with clinicopathological characteristics. Methods The expressions of MMP-2 and CXCR4 mRNA in 68 cases of fresh ccRCC tissues and 36 cases of normal renal tissues were detected by reverse transcription polymerase chain reaction (RT-PCR). Re- suits The half-quantity values of MMP-2 and CXCR4 mRNA in ccRCC tissues were 0.848+_0.357 and 0. 939+0. 336,which were significantly higher than those in normal tissues (t = 7. 375,8. 860, P^0.05). Furthermore, in ccRCC tissues the expres- sions of MMP-2 and CXCR4 mRNA had a significant association with clinical stage,histological grade and lymph node metasta- sis (t =2. 169--3. 852,P-~0.05) ,while not with gender and age of patients and the diameter of tumors (t=0.201t0.630,P~〉 0.05). Moreover, there was a positive correlation between the expression of MMP-2 and CXCR4 mRNA in human ccRCC tis- sues (r=0.832,P^0.05). Oonclusions These results suggested that MMP-2 andCXCR4 might play a cooperative role in the growth,invasion and metastasis of ccRCC. The co-detection of MMP-2 and CXCR4 could be conducive to the diagnosis and evaluation of ccRCC.
