线粒体基因7.4kbp大片段缺失突变与侵袭性牙周炎的关系研究 被引量:1

摘要 目的:探讨线粒体基因(mtDNA)7.4 kbp大片段缺失突变和侵袭性牙周炎(AgP)的关系。方法:选取20例AgP患者(AgP组)和20例牙周健康者(对照组),收集外周血及牙周翻瓣术及牙冠延长术中切取的牙龈组织,同时收集10例慢性牙周炎者(CP组)牙周翻瓣术中切取的组织。采用长距离PCR方法检测血样本和牙龈组织样本中mtDNA 7.4 kbp大片段缺失,比较3组间的差异。结果:AgP组20例牙龈组织中1例存在mtDNA 7.4 kbp大片段缺失,AgP组2例组织和CP组1例组织中检测到目前尚未报告过的mtDNA 5 537 bp大片段缺失突变。所有血样和对照者牙龈组织中均未检测到大片段缺失突变。结论:AgP患者牙龈组织中存在mtDNA 7.4 kbp大片段缺失,发现一种新的mtDNA 5 537 bp大片段缺失突变。 Objective: To study the mitochondria DNA(mtDNA) 7.4 kbp deletion in patients with aggressive periodontitis(AgP). Methods: Blood and gingival tissue samples were collected from 20 patients with AgP( AgP group), 20 healthy control subjects ( Control group) and gingival tissue from 10 patients with chronic periodontitis( CP group). Long range PCR was used to detect mtD- NA 7.4 kbp deletion in all subjects. Results: MtDNA 7.4 kbp deletion was found in gingival tissue of 1 patient with AgP, a novel mtDNA 5537bp deletion was detected in 2 cases of AgP group and 1 of CP group. No deletion was found in the blood samples of all subjects and in gingival tissue in control subjects. Conclusion: MtDNA 7.4 kbp deletion and a novel mtDNA 5 537 bp deletion in gingival tissue may be related with aggressive periodontitis.
