摘要
目的观察利塞膦酸钠联合脉冲电磁场对去势大鼠骨质疏松的疗效。方法将32只雌性SD大鼠(250±20)g随机分成4组:正常生长组(Sham)、去卵巢模型组(OVX)、利塞膦酸钠治疗组(RIS)和利塞膦酸钠联合骨质疏松治疗仪治疗组(RIS+PEMFs),每组8只。后三组大鼠行双侧卵巢切除术。去势后12 w RIS组给予0.1 mg·kg-1·d-1利塞膦酸钠灌胃治疗,RIS+PEMFs组给予0.1 mg·kg-1·d-1利塞膦酸钠灌胃和1h·d-1脉冲电磁场治疗,Sham、OVX组每天给予等量生理盐水灌胃。去势后12 w、灌胃治疗4 w、8 w、12 w后四组大鼠均用双能X线吸收法骨密度仪测量全身骨密度(BMD),治疗12 w后进行血生化检测、骨CT值测量和骨组织形态学观察。结果去势后12w后三组BMD与Sham组比较均显著降低(P<0.01),大鼠骨质疏松模型确立;灌胃治疗12 w后,RIS、RIS+PEMFs两组较OVX组BMD(P<0.01)、骨CT值(P<0.01)均较高,且RIS+PEMFs组BMD、骨CT值均比RIS组高(P<0.01);RIS组血清Ca、ALP浓度均较RIS+PEMFs、Sham组高(P<0.05),且RIS+PEMFs组血清ALP浓度较Sham组相比差异无统计学意义(P>0.05);股骨颈病理形态学观察中RIS+PEMFs组的骨小梁形态较完整,较接近Sham组水平。结论去势后12 w可建立大鼠骨质疏松模型;利塞膦酸钠对去势所致骨质疏松大鼠有治疗作用,其与脉冲电磁场联合的治疗效果更为显著。
Objective To observe the clinical efficacy of risedronate combined with pulsed electromagnetic fields ( PEMFs) for the treatment of osteoporosis in ovariectomized rats.Methods Thirty-two female SD rats were randomly divided into 4 groups:sham group (Sham), OVX group (OVX), risedronate group (RIS), and risedronate combined with PEMFs group (RIS+PEMFs). Each group had 8 rats.Rats in the last 3 groups received the operation of bilateral ovariectomy.At the 12 th week after the operation, rats in RIS group were given a gavage of 0.1mg kg-1 d-1 risedronate, and rats in RIS +PEMFs group were received a gavage of 0.1mg kg-1 d-1 risedronate and 1h d-1 pulsed electromagnetic field therapy.Rats in Sham group and OVX group were given a gavage of same volume of normal saline.Bone mineral density ( BMD) of the total body was detected using dual energy X-ray absorptiometry at the 12thweek after ovariectomy and the 4th week, the 8th week, and the 12thweek after the gavage.After 12-week treatment, the bone biochemical detection, bone CT detection, and the histomorphological observation of bone tissues were performed.Results At the 12 th week after ovariectomy, BMD in the last 3 group was significantly lower than that in Sham group (P&lt;0.01), indicating the establishment of rat osteoporosis model.After 12-week gavage, BMD in RIS group and RIS+PEMFs group and the values of bone CT were higher than those in OVX group (P&lt;0.01), and the values in RIS+PEMFs group were also&amp;nbsp;higher than those in RIS group (P&lt;0.01).The serum concentrations of Ca and ALP in RIS group were higher than those in Sham group and RIS+PEMFs group (P&lt;0.05), while no significant difference of serum ALP between RIS+PEMFs group and Sham group was observed ( P&gt;0.05 ) .The pathological morphology observation of the femoral neck showed that the bone trabecula in RIS+PEMFs group was intact, closed to the level in Sham group.Conclusion The rat osteoporosis model can be established at the 12thweek after ovariectomy.Risedronate has therapeutic effect on osteoporosis in overiectomized rats , and the effect is more obvious when combined with PEMFs .
