摘要
目的探讨CRP在小鼠肺炎支原体肺炎模型中的表达及相关作用机制。方法建立小鼠肺炎支原体肺炎模型,分为对照组、模型组、治疗组,每组8只,按时间点处死后留取血清、肺及其他脏器组织,应用酶联免疫法、免疫组织化学技术及RT-PCR方法检测对照组、模型组、治疗组CRP的表达。采用SPSS11.5软件包进行统计学处理。结果CRP在肺炎支原体肺炎的肺组织中呈高表达,与正常组、治疗组比较差异具有显著性(P<0.05)。CRP在有肺外炎症的阳性表达强度明显高于无肺外炎症的组织中的表达,差异有显著性(P<0.05)。酶联免疫法、免疫组织化学检测CRP蛋白阳性表达结果与RT-PCR的方法检测结果一致。结论 CRP在肺炎支原体肺炎中的表达高于其在正常组、治疗组中的表达;其阳性表达强度与肺炎支原体肺炎的炎症程度有关。CRP在有肺外炎症的阳性表达强度明显高于无肺外炎症的组织中的表达。CRP有可能参与了肺炎支原体肺炎的发生发展过程,为其诊断、肺外炎症的提示及合理治疗提供依据。
Objective To research CRP expression in MPP,evaluate there clinical significance and mechanism. Meth- ods A mouse model of mycoplasma pneumonia pneumonia, control group, model group, treatment group each consis- ting of eight mice were established. Saving blood serum, lungs and other visceral organ, We detected the expression of CRP in control group, model group and treatment group by ELISA, immunohistochemistry and RT-PCR. Statistical treatment was carried out with SPSSll. 5 software package. Results CRP have a high expression in lung tissue of model group which is significantly different with the expression in control group and treatment group (P〈0.05). CRP have an apparent higher positive expression in the mice that has extrapulmonary complication than doesn't has it(P〈0.05). With the detection of ELISA and immunohistochemistry CRP provided positive results. And it was highly consistent with RT-PCR. Conclusion CRP have a significantly higher expression in model group lung tissue than in control group and treatment group. Its expression intensity is related to the severity of MPP. CRP have significantly high expressions in the mice with extrapulmonary complication than those without. CRP might be involved in the development and pro- gression of MPP. Provide theory and experiment basis for MPP clinical diagnosis,prompting extrapulmonary complica- tion and treatment.
