少腹逐瘀汤对寒凝血瘀模型大鼠红细胞膜组分的影响 被引量:15

摘要 目的从血液流变学和红细胞膜组分的变化探讨少腹逐瘀汤改善寒凝血瘀模型大鼠血瘀证的作用机制。方法 40只Wistar大鼠,随机分为空白组、寒凝血瘀模型组、少腹逐瘀汤高剂量组、少腹逐瘀汤低剂量组,每组10只。大鼠置于0℃~1℃冰水中8~10min,待全身僵直后取出,连续造模14天。给药组每日给予相应药物1次。第15天大鼠皮下注射盐酸肾上腺素0.8μl/g(0.8μg/g),g.2次,间隔4h,2次之间将大鼠置于冰水中浸泡5min,末次造模后禁食不禁水12h。取血测定血液流变学指标以及红细胞膜组分相关指标。结果与空白组比较,寒凝血瘀模型组大鼠全血黏度升高,红细胞变形指数降低,FIB和APTT升高,Na+-K+-ATP酶、SOD活力及唾液酸含量降低,膜胆固醇含量升高,差异均有统计学意义(P〈0.05,P〈O.01)。与寒凝血瘀模型组比较,少腹逐瘀汤能够降低全血黏度,升高600S。和800S。切变率下红细胞变形指数,延长AVFT和1Tr,升高血浆TG含量,并可升高唾液酸含量、Na+-K+-ATP酶及SOD活力,降低膜MDA和血浆FIB含量,差异均有统计学意义(P〈O.05,P〈0.01)。结论少腹逐瘀汤可改善寒凝血瘀模型大鼠血液的高凝状态,在减轻自由基对红细胞攻击程度、抗氧化,以及提高红细胞膜Na+-K+-ATP酶的活性,维持细胞正常形态方面具有一定优势。 Objective To investigate the effect of "Shaofu Zhuyu Decoction" (SFZYD) on blood stasis pattern in rats and to discuss its mechniasm based on the hemorheology and components of erythrocyte membrane. Methods Forty Wistar rats were randomly assigned to blank group, model group and SFZYD high-dose group and low-dose group with l0 rats in each group. These rats were put into icewater (0℃~1℃ ) for 8-10 minutes until the rats were titanic everyday. The SFZYD were given everyday and this method was used for 14 days. In the 15th day 0.8 ml. kg-/ adrenalin hydroehloride was injected into rats with hype for twice which interval 4 hours. The related indexes of hemorheology and the components of erythrocyte membrane were determined. Results Compared with the model group, the whole blood viscosity (P 〈0.01 ) was decreased and the red blood cell deformability under 600 s -I and 800 s-1 were increased (P 〈0.01 ) by SFZYD. APTT and TY (P 〈0.05, P 〈0.05 ) were extended by SFZYD. The content of TG in plasma was increased (P 〈0.05 ). The content of sialie acid ( P 〈0.05 ), Na + -K + -ATP enzyme and SOD (P 〈0.05, P 〈0.05 ) in erythrocyte membrane were increased by SFZYD. The content of MDA ( P 〈 0.05 ) in erythrocyte membrane and FIB in plasma ( P 〈0.05 ) were decreased by SFZYD. Conclusion SFZYD could improve the situation of blood stasis through reducing the damage of free radical on red blood cell and oxidation resistance. And SFZYD has the advantage in maintaining cell morphology.
