摘要 目的:建立了微流控芯片毛细管电泳激光诱导荧光检测法检测白屈菜中血根碱的方法.方法:以pH-9.0的20 mmol/L硼砂-SDS为电泳缓冲溶液,荧光素钠为内标物,分离电压1.8 kV,在微流控芯片上检测白屈菜中的血根碱.分别考察了缓冲溶液浓度、pH、添加剂、分离电压对检测的影响.结果:SDS的加入可使血根碱的荧光增敏,80 s内可完成血根碱和荧光素钠的分离.血根碱的线性范围为99.6~99.7μg/L(r=0.9990).结论:此方法灵敏、快速、准确,可用于检测白屈菜中的血根碱. Objection: To develop a method for the determination of sanguinarine in Chelidonim majus by microchip capillary electrophoresis with laser induced fluorescence detector. Methods: Sanguinarine was separated with fluorescein sodium as an internal standard on microchips in 20 rnmol/L borate-SDS buffer solution under 1.8 kV. The concentration and pH value of buffer solution, the additives and the separation voltage was optimized. Results: Fluorescence of sanguinarine was increased by the addition of SDS. Sanguinarine and fluoresecein sodium can be separated in 80 s. The linear range of sanguinarine was 99.6-99.7 μg/L (r=0.9990). Conclusion: This method is simple, sensitive, accurate, and it could be used for the determination of sanguinarine.
