摘要
目的 分析全反式维甲酸及中药香龙散含药血清对胃癌细胞端粒酶活性的影响 ,并探讨端粒酶活性和细胞凋亡的关系。方法 用纯种新西兰家兔制备中药香龙散含药血清 ,应用TRAP分析全反式维甲酸及中药香龙散含药血清对培养的胃癌细胞端粒酶活性的影响 ,应用DNALadder分析及流式细胞仪观察全反式维甲酸及中药香龙散含药血清对培养的胃癌细胞凋亡的影响。结果 首先全反式维甲酸和中药香龙散含药血清对培养的胃癌细胞生长的抑制有协同作用。其次 ,全反式维甲酸处理胃癌细胞 2 4小时 ,即可见端粒酶活性下调 ,中药香龙散含药血清无此作用 ,也不能加强全反式维甲酸对端粒酶活性下调的作用。最后 ,在全反式维甲酸处理的胃癌细胞中 ,第 3天出现了细胞凋亡 ,并且中药香龙散含药血清对全反式维甲酸诱导的细胞凋亡有加强作用。结论 全反式维甲酸或 /和中药香龙散含药血清对培养的胃癌细胞生长有抑制作用 ,其中全反式维甲酸可能通过抑制端粒酶活性表达而诱导了胃癌细胞凋亡 ,中药香龙散含药血清有加强全反式维甲酸诱导细胞凋亡的作用 。
Aim To study the effect of ATRA(all-trans retinoic acid) and serum with Xianglong pulvis on the telomerase activity in the gastric carcinoma cell line, and explore the relationship between telomerase activity and apoptosis, in order to find out inhibitor of telomerase and open up the new way of cancer therapy. Method The serums with Xianglong pulvis were prepared using pure breed New Zealand rabbits. The effect of ATRA and serum with Xianglong pulvis on the telomerase activity and the apoptosis of the cultured gastric carcinoma cells were studied by TRAP( telomere repeat amplification protocol), DNA ladder as well as flow cytometry. Result The inhibition of growth of cultrued gastric carcinoma cells (HS746T) differs depending on the concentration of ATRA or/and serum with Xianglong pulvis. Further, 10μmol/L of ATRA or/and 10% of serum with Xianglong pulvis was an optimum concentration. Both of them cooperated to inhibit the growth of cultured gastric carcinoma cells. In addition, the telomerase activity had been partially decreased at 10 μmol/L ATRA for 1 day and completely inhibited for 5 days, but serum with Xianglong pulvis can neither decrease the telomerase activity itself nor enhance the ATRA′s effect. Moreover, the cellular apoptosis was observed at third day when ATRA was 10μmol/L. 10% serum with Xianglong pulvis enhanced cellular apoptosis induced by ATRA. Conclusion The ATRA or/and serum with Xianglong pulvis can inhibit the growth of cultured gastric cancer cells. The ATRA can induce cellular apoptosis by inhibiting telomerase activity. The serum with Xianglong pulvis can enhance cellular apoptosis induced by ATRA, but the mechanism is unknown.
