羟基红花黄色素A对小鼠Lewis肺癌移植瘤的抑制作用及VEGF表达的影响 被引量:12

摘要 目的观察羟基红花黄色素A(hydroxysafflor yellow A,HSYA)对C57BL/6小鼠Lewis肺癌移植瘤的抑制作用,并观察其对肿瘤组织血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法 C57BL/6近交系小鼠30只,造模后,分为5组,每组6只,分别使用生理盐水、环磷酰胺(CTX)及HSYA(分小、中、大3个剂量)作用于动物模型。观察各组肿瘤体积大小,并在造模第22天处死所有小鼠,无菌条件下剥取瘤组织并称重,计算抑瘤率;应用免疫组化法及图像分析技术检测瘤组织VEGF表达的情况。结果 HSYA中、小剂量组,在瘤株接种第10、16、22天肿瘤体积与生理盐水组比较差异有统计学意义(P<0.05);在第22天,瘤重与生理盐水组比较差异有统计学意义(P<0.05);在肿瘤组织中VEGF表达上,中、小剂量组与生理盐水组比较差异有统计学意义(P<0.05),而HSYA大剂量组肿瘤体积、瘤重及VEGF表达上均与生理盐水组比较差异无统计学意义。结论羟基红花黄色素A能抑制Lewis肺癌小鼠肿瘤生长,并能降低小鼠Lewis肺癌移植瘤组织中VEGF的表达。 Objective To study the inhibitive effect of safflor yellow A on transplanted tumor of Lewis lung cancer and vascular endothelial growth factor (VEGF) expression in C57BL/6 mice. Methods The model of C57 BIV6 mice bearing Lewis lung cancer was established by subcutaneous seeding of Lewis lung cancer cells, and randomly divided into 5 groups (n =6). Tumorbearing mice were injected with normal saline, cyclophosphamide (CTX) or safflor yellow A ( low, medium or high dose) in each group. The tumor volume was measured. All mice were sacrificed on Day 22nd under aseptic condition for the tumor collection. The transplanted tumors were weighed for calculation of the tumor inhibition rate. hnmunohistochemical method and image analysis technique were applied to assessing the VEGF expression in tunmr tissue. Results In small dose and medium dose HSYA groups, tumor volume were significantly smaller than that in con- trol group on Day 14th, 16th and 22nd(P 〈 0. 05 ). There were significant differences in tumor weight and VEGF expression between in small dose or medium dose HSYA group and control group (P 〈 0. 05 ). However, there was no significant dit'ference in tumor volume, tumor weight or VEGF expression between high dose HSYA group and control group. Conclusion HSYA can inhibit tumor growth of Lewis lung cancer in mice, also down - regulates the expression of VEGF.
