摘要
蚯蚓在DMP质量浓度为200mg/kg和600mg/kg的自然土壤中污染胁迫7,14,21d,采用蛋白质双向电泳技术筛选出表皮组织差异表达的140个蛋白质斑点,并用肽质量指纹图谱技术和数据库比对法初步鉴定出5种差异蛋白质,分别为假定蛋白(BRAFLDRAFT_58397),α微管蛋白,ATP合酶β亚基,β微管蛋白,假定蛋白(FAEPRAM212_03570),这些蛋白质可能参与邻苯二甲酸二甲酯代谢并与蚯蚓解毒功能有关,有望成为土壤生态毒性评价、监测和开展毒理学研究的生物标记物.
Extracts of whole earthworms,living in a DMP-polluted environment (200 mg/kg,600 mg/kg),collected at days 7,14,and 21 were analyzed by 2-DE and quantitative image analysis.In total,140 proteins demonstrated a signifcant regulation for at least one time point.Compared with control samples,part of differential protein spots were identified by peptide mass fingerprint based on MOLDI-TOF-MS.Five differential expressed proteins were identified,including BRAFLDRAFT_58397,Alpha tubulin,ATP synthase beta subunit,Beta tubulin,FAEPRAM212_03570.These proteins were involved in several processes,including metabolism and detoxification function.We suggest that the variable levels and trends in these spots on the gel may be useful as biomarker profiles to investigate DMP contamination levels in soils.
