注射性坐骨神经损伤后针刺对家兔腓肠肌内NADH-TR和胶原纤维的影响 被引量:5

Effects of acupuncture on gastrocnemius muscle after sciatic nerve injection injury in rabbits

摘要 目的探索注射性坐骨神经损伤后针刺腧穴改善肌萎缩的机制。方法成年家兔42只,随机分为7组:正常组;青霉素注射坐骨神经2周和4周组;青霉素注射坐骨神经加针刺腧穴4周和8周组;青霉素注射坐骨神经加针刺尾部对照4周和8周组。取腓肠肌外侧头横切片作烟酰胺腺嘌呤二核苷酸四唑氧化还原酶染色与VG染色。结果青霉素注射2周时,腓肠肌外侧头内烟酰胺腺嘌呤二核苷酸四唑氧化还原酶染色加深,光密度值增大,肌纤维横切面积减小,肌重减轻,胶原纤维增多。I型和II型纤维的比例不变,IIA型纤维比例增加,IIB型纤维相应减少。4周时上述变化更重。坐骨神经注射性损伤4周后,针刺穴位治疗4周,上述变化明显恢复。8周时,进一步好转。尾部针刺对照组无恢复迹象。结论针刺腧穴改善注射性坐骨神经损伤所致的肌萎缩可通过降低肌内烟酰胺腺嘌呤二核苷酸四唑氧化还原酶活性和减少胶原纤维增生实现。 Objective To investigate the mechanism of acupuncture to improve muscle atrophy caused by injec- tion- induced sciatic nerve injuries. Methods 42 adult rabbits were randomly divided into 7 groups: normal; penicillin injection 2 and 4 weeks groups; acupunctured 4 and 8 weeks groups; acupuncture on tail 4 and 8 weeks control groups. The transverse section of the lateral head of the gastrocnemius muscle was stained by nico- tinamide adenine dinucleotide four tetrazolium oxidoreduetase and VG staining methods. Results Injection of penicillin for 2 weeks resulted in deepened staining of nieotinamide adenine dinucleotide tetrazolium oxidoreduc- tase, increased value of optical density, decreased cross sectional area of muscle fibers, lightened muscle weight, and increased collagen fibers in cross sections of lateral head of gastrocnemius muscle. The ratio of type I and type Ⅱ fibers is constant, with an increased proportion of type IIA fibers and decreased type B fibers. The above changes were worse at 4 weeks. In sciatic nerve injection injuries at Ⅱ4 weeks together with acupuncture treatment for 4 weeks, the above changes were obviously recovered. At 8 weeks, those changes were further im- proved. No signs of recovery were observed in the control group of acupuncture on tail. Conclusion Acupuncture improves muscle atrophy caused by sciatic nerve injection injuries. This effect appears to be mediated by decrea- sing the activity of nicotinamide adenine dinucleotide tetrazolium oxidoreductase and reducing the hyperplasia of collagen fibers in muscles.
