穴位埋线对VD大鼠学习记忆能力及血清Livin表达的影响 被引量:1

摘要 目的观察穴位埋线对血管性痴呆(vascular dementia,VD)大鼠学习记忆能力和血清抗凋亡因子(Livin)含量的影响,探讨穴位埋线对VD大鼠脑缺血性损伤的神经保护机制。方法将大鼠随机分成假手术组、VD模型组、穴位埋线组、尼莫地平组,采用改良Pulsinelli’s四血管闭塞法(four-vessel occlusion,4-VO)建立VD大鼠模型,2个治疗组分别施行穴位埋线和尼莫地平治疗,连续15d,Morris水迷宫检测各组大鼠学习记忆能力,酶联免疫吸附法测定血清Livin含量,制作海马组织病理切片并进行HE染色,观察并比较各组大鼠学习能力、血清Livin含量及海马神经元损伤的变化。结果模型组表现出明显的学习记忆障碍,定位航行试验、空间探索试验与假手术组相比有显著性差异(P<0.01);与VD模型组比较,穴位埋线治疗后VD大鼠学习记忆能力显著提高(P<0.05,P<0.01),血清Livin含量升高(P<0.05)。结论穴位埋线可改善VD大鼠学习记忆能力,其机理可能与埋线后提升血清Livin含量,进一步抑制神经细胞凋亡有关。 Objective: To observe the effect of acupoint catgut implantation on the learning ability and memory of vascular dementia( VD) rats and the influence of serum anti- apoptotic factors( Livin) content and to explore the neuroprotection mechanism of the acupoint catgut implantation for cerebral ischemic injury of VD rats. Methods: The rats were randomly divided into a sham- operation group,a VD model group,an acupoint catgut implantation group and a nimodipine group. A modified Pulsinelli's four- vessel occlusion( 4- VO) was used to make VD rat model. Two treatment groups were respectively treated with the acupoint catgut implantation and nimodipine for 15 days and Morris water maze test was used to measure the learning ability and memory of the rats in each group. An enzyme linked immunosorbent assay was used to determine the serum Livin content and hippocampal tissue pathological slices was made for HE staining to compare and observe the learning ability and Livin serum content and hippocampal neuronal damage changes of theVD rats. Results: The model group showed significant learning and memory impairment. When compared with the sham- operation group,location navigation test and space exploration test showed a significant difference( P < 0. 01). When compared with the VD model group,the learning ability and memory of the acupoint catgut implantation group significantly improved after the implantation with serum Livin content increased( P < 0. 05). Conclusion: The acupoint catgut implantation can improve the learning ability and memory of VD rats,which may be related to the increase of the serum Livin content after the implantation and the inhibition of neuronal apoptosis.
