摘要
目的 :观察银翘玉屏风汤拮抗急性风寒应激小鼠免疫功能的抑制作用。方法 :昆明种小鼠随机分成 5组 ,即正常对照组(N)、模型组 (M)、玉屏风组 (YPF)、高剂量银翘玉屏风汤组 (HYQ YPF)、低剂量银翘玉屏风汤组 (L YQYPF)。小鼠年龄 6~ 8周 ,雌雄各半 ,每组小鼠 7~ 9只 ,实验前饲养 3d,每组动物灌胃给予相同体积的凉开水或药物。 1周后 ,按照改进的陈新 [1 ]的方法给小鼠风寒刺激。3 d后处死小鼠 ,开始检测各项指标。IL -2含量测定用 RIA法 ,同时检测了胸腺指数和观察了胸腺的微观结构。结果 :应激小鼠的胸腺指数明显降低 (P<0 .0 1) ,胸腺发生急性病理性萎缩 ,IL-2含量、腹腔巨噬细胞吞噬功能、外周血淋巴细胞对 PHA的增殖反应均明显下降。 YPF ,H YQYPF ,L YQYPF对应激小鼠的免疫功能均有保护作用 ,其中以 HYQYPF ,L YQYPF拮抗应激小鼠免疫功能抑制的作用最明显。结论 :急性风寒应激介导的免疫抑制是由于激活了下丘脑 -垂体 -肾上腺轴 ;YPF,HYQYPF,L YQYPF均能拮抗应激小鼠免疫功能的抑制 ,其中以 YQYPF(尤其 HYQ YPF)
Objective: To observe the antagonistic effect of Yinqiao Yupingfeng decoction on the immunosuppression in acute wind-cold stressed mice. Methods:[Kunming mice were divided into 5 groups at random: normal group (N), model group (M), Yupingfeng granules (YPF), high dosage Yinqiao Yupingfeng decoction (HYQYPF), low dosage Yinqiao Yupingfeng (LYQYPF). The animals were used in age of 8 to 10 weeks. The number of female mice was equal to that of male mice. Each group consisted of 7 to 9 mice. Mice had been bred and housed for 3 days before the beginning of every experiment. Each mouse was fed same volume boiled water or decoction by intragastric injection. A week later, the mice of model group and treatment groups were stressed by cold fluid mixture of water and ice (0±1)℃ for 8 minutes. Then the mice were placed in a wind-cold stress box for 3 hours. 3 days later, the mice were killed and immunologic function was assayed. Radio immunoassay (RIA) was used for plasma IL-2 concentration. The thymic weight index was calculated and the thymic microscopic structure was studied also. [WT5”HZ]Results: The thymic weight index of stressed mice decreased obviously (P<0.001). There was acute pathological atrophy in the thyms of stressed mice. The serum IL-2 level, the peritoneal exudate macrophage function, the responsiveness of peripheral lymphocyte to PHA decreased significantly. YPF, HYQYPF, LYQYPF all had protective effect on the immunologic function. Among the three decoctions, HYQYPF, LYQYPF showed the most obvious antagonist to immunosuppression. Conclusion: The immunosuppression caused by acute wind-cold stress is due to activating the pothalamopituitary-adrenal axis. YPF, HYQYPF, LYQYPF all could antagonize the immunosuppression of stressed mice. Among these, YQYPF (above all HYQYPF) has the most significant effect on the enhancement of immunologic function.
