大鼠膈肌水溶性蛋白组的研究

摘要 本研究应用蛋白组的核心技术———高分辨率二维电泳对同属骨骼肌的膈肌和趾长伸肌水溶性蛋白组的二维电泳图谱进行了比较 ,并对两者在老化过程与年龄相关的水溶性蛋白进行了比较。我们发现 :膈肌和趾长伸肌水溶性蛋白的二维图谱极为相似 ,大部分蛋白都能较好地重叠。我们未能找到持续而稳定且特异于膈肌的水溶性蛋白。同时我们发现了三个与年龄相关蛋白 (S1,S2和S3) ,随增龄 ,蛋白S1只在 2 4月龄趾长伸肌存在 ,而在 8月龄和 2 4月龄的膈肌中却不能被检测到。S2在 8月龄的膈肌和趾长伸肌存在 ,而在 2 4月龄的膈肌和趾长伸肌中不能被检测到。S3在 8月龄的膈肌和趾长伸肌中不能被检测 ,而在 2 4月龄的膈肌和趾长伸肌中能被检测到。本研究提示 :膈肌和趾长伸肌水溶性蛋白在其成年时表达基本一致。 The aqueous proteome of extensor digitorum longus (EDL) muscle and diaphragm (DIA) muscle were established by high resolution two-dimensional electrophoresis (2-DE).The differences between the 2-DE patterns obtained from EDL and DIA muscles were compared.It was found that the overall 2-DE patterns of these two muscles are similar.Most of resolved proteins in the 2-DE maps of EDL and DIA muscles were well matched and could be superimposed.We could not find a representative protein that was unique for DIA msucle.In addition, three age-related proteins (which we named S1, S2 and S3) were screened.It was found that S1 could only be detected in old (24 month-old) EDL muscles, but in the young (8 month-old) and old DIA muscles.One endogenous protein, S2, became undetectable in old EDL and DIA muscles.S3 was not present in young muscles but its expression was induced in old EDL and DIA muscles.Our findings suggested that the 2-DE aqueous protein profiles of EDL and DIA muscles are similar.The age-related changes of aqueous proteins in these two muscles are also similar.
