细胞凋亡抑制蛋白IAPs的研究进展与系统进化分析 被引量:2

摘要 凋亡抑制蛋白(inhibitors of apotosis protein,IAP)是细胞凋亡过程中的重要调控因子,具有抑制细胞凋亡的作用。本文综述国内外学者在IAPs蛋白结构和功能及家族新成员鉴定等方面的研究成果,重点介绍了病毒、酵母、线虫、果蝇、家蚕等的IAPs蛋白的3个主要结构域———杆状病毒IAP重复序列结构域(BIR)、RING锌指结构域、半胱天冬酶活化募集结构域(CARD),以及IAPs在抑制半胱天冬酶Caspase3/7/9催化活性和阻断细胞凋亡进程中的生物学功能。对IAPs家族进行系统进化分析,推测2种核型多角体病毒(云杉卷叶蛾核型多角体病毒和油桐尺蠖核型多角体病毒)在适应性进化过程中获得了与宿主细胞IAPs蛋白结构与功能类似的IAPs蛋白。 Inhibitors of apoptosis protein (lAP) is an important regulatory factor with anti-apoptotic function in the process of cell apoptosis. In this article, we reviewed the domestic and overseas research achievements on lAPs including protein structure and function, and identification of new family members. We mainly analyzed three major domains of lAPs proteins in virus, yeast, Caenorhabditis elegans, Drosophila melanogaster and Bombyx mori. The three domains are repetitive sequence of lAPs in baculovirus (BIR), RING zinc finger domain and Caspase-activated recruitment domain (CARD). Besides, the biological function of lAPs in inhibiting Caspase3/7/9 catalytic activity and blocking apoptosis process was discussed. Moreover, we conducted phylogenetic analysis of lAPs family members, and inferred that Choristoneura fumifierana nucleopolyhedrovirus and Buzura supressaria nucleopolyhedrovirus acquired lAPs protein with the similar protein structure and function as host cells in the process of adaptive evolution.
