摘要
目的 :研究大肠癌患者外周血中 CK 2 0 m RNA作为大肠癌细胞微转移的标志。方法 :巢式逆转录多聚酶反应 (RT- PCR)法。结果 :CK2 0 m RNA巢式 RT- PCR最低可检测到每 5 ml血液中仅存 5个结肠癌细胞的存在 ;大肠癌组 32例 ,外周血中 CK 2 0 m RNA阳性率 5 6 .2 6 % (18/32 ) ;对照组中 CK 2 0 m RNA均为阴性。结论 :CK 2 0m RNA巢式 RT- PCR可作为大肠癌细胞微转移的敏感而特异的检测方法。
Objective:To indicate whether the expression of CK20mRNA in peripheral blood can be a helpful marker of micrometastases of colorectal carcinomas. Method:Nested reverse transcriptase-ploymerase chain reaction. Results:The detection of CK20mRNA in peripheral blood by nested reverse transcriptase-ploymerase chain reaction (RT-PCR) was sensitive enough to detect colorectal carcinoma cells, the limit of identification was 5 carcinoma cells in every 5 millilitres. The expression of CK20mRNA in peripheral blood of 10 cases of contral group were all negative . The positive rate of CK20mRNA in peripheral blood of total 32 patients with colorectal carcinomas was 56.26%(18/32). Conclusion:Nested reverse transcriptase-ploymerase chain reaction was a sensitive and specific methold to indicate the expression of CK20mRNA in peripheral blood as a helpful maker of micrometastases of colorectal carcinoma cells.
