摘要
目的:研究人肠内细菌对牛蒡苷48 h的转化和转化酶的性质。方法:采用人肠内细菌的混合菌液与牛蒡苷在37℃厌氧条件下进行培养;采用硅胶柱色谱和制备HPLC分离转化产物;根据核磁共振波谱鉴定转化产物的结构;采用超声破碎方法,提取人肠内细菌总酶。结果:牛蒡苷在48 h内可被人肠内细菌转化为牛蒡苷元和牛蒡苷元脱甲基化产物;转化酶可能为组成酶和诱导酶。结论:牛蒡苷在48 h时主要转化产物为牛蒡苷元和脱甲基化产物。
Objective: To study the biotransformation of arctiin by human intestinal flora (HIF) within 48 h and the characteristics of transformation enzymes. Methods: The arctiin was anaerobically incubated with HIF at 37℃ for 48 h. The biotransformation products were isolated and purified by silica gel column chromatography and prepared HPLC. Chemical structures of the transformed products were determined by nuclear magnetic resonance (NMR) method. The enzymes of HIF were extracted by uhrasonication. Results: Arctiin was converted to arctigenin and 3-demethylarctigenin. Its transform enzymes were probably constitutive enzymes and induced enzymes. Conclusion: The main biotransformation products of arctiin were arctigenin and 3-demethylarctigenin within 48 h.
