摘要
目的研究同型半胱氨酸(Hcy)对人脐静脉内皮细胞总甲基化水平的影响。方法体外培养人脐静脉内皮细胞,并经Ⅷ因子法对其进行鉴定,分别用含有0、2、8、32 mmol·L-1Hcy的RPMI 1640培养液对其作用48 h后,测定基因组总甲基化水平。结果 0、2、8、32 mmol·L-1Hcy组HUVEC细胞总甲基化水平分别为:(9.25±0.15)%、(8.53±0.49)%、(6.24±0.27)%、(3.79±0.13)%,呈明显的剂量效应关系(P<0.05)。结论 Hcy可下调人脐静脉内皮细胞DNA的总甲基化水平,并可能是Hcy致动脉粥样硬化的重要机制之一。
Objective To study the effects of homocysteine on global methylation level of human umbilical vein endothelial cell. Methods The human umbilical vein endothelial cells (HUVEC) were cultured in vitro and indentified by Factor Ⅷ Related Anti- gen. After treated with 0, 2, 8 and 32 mmol L- 1 Hcy for 48h, Methylamp TMGlobal DNA Methylation Quantification Kit was used to detect the level of DNA methylation. Results Comparing with the control group, the rate of global DNA methylation in 0, 2, 8 and 32 mmol L- 1 Hcy group were respectively (9.25 ± 0.15 ) %, ( 8.53 ± 0.49) %, (6.24 ± 0.27 ) % and ( 3.79± 0.13 ) % , and presented in a dose - dependant manner ( P 〈 0.05 ).
