摘要
目的:评价调补肺肾(补肺健脾、补肺益肾、益气滋肾)三法对慢性阻塞性肺疾病(COPD)大鼠肺脏氧化应激的影响。方法:大鼠随机分为对照组、模型组、补肺健脾组、补肺益肾组、益气滋肾组和氨茶碱组,采用香烟暴露联合反复细菌感染法制备COPD大鼠模型。第9-20周对照组、模型组给予0.9%氯化钠溶液,其余组分别给予相应药物灌胃,于第20、32周观察肺功能,前列腺素E2(PGE2)、超氧化物歧化酶3(SOD3)mRNA及磷酸化过氧化体增殖物激活受体γ(p-PPARγ)蛋白表达。结果:第20、32周,模型组大鼠肺功能(VT、PEF、EF50)、肺组织SOD3及p-PPARγ表达较对照组降低(P<0.01),PGE2升高(P<0.01);3个中药组及氨茶碱组除PGE2外均能够改善上述指标变化,3个中药组优于氨茶碱。结论:调补肺肾三法和氨茶碱均能不同程度改善COPD氧化应激状态,调补肺肾三法作用优于氨茶碱,其中以补肺益肾方作用尤为明显。
Objectives: To evaluate the influence and long-term effect of the three tiaobu feishen therapies (bufei jianpi, bufei yishen and yiqi zishen) of traditional Chinese medicine (TCM) on oxidative stress in rats with chronic obstructive pulmonary disease (COPD). Methods: Rats were randomly divided into control group, model group, bufei jianpi group, bufei yishen group, yiqi zishen group and aminophyline group. COPD rat model was duplicated by smoke inhalation and bacterial infections. From the 9th to 20th week, the COPD rats were administrated with normal saline (in control and model group), Bufei Jianpi Granule (in bufei jianpi group), Bufei Yishen Granule (in bufei yishen group), Yiqi Zishen Granule (in yiqi zishen group) and Aminophyline (in aminophyline group). At the 20th and 32nd week, half of the animals were sacrificed, and expressions of PGE2 and SOD3 mRNA, and PPARγ were detected. Results: At the 20th and 32nd week, lung function (Vx, PEF and EF50 ), SOD3 mRNA and p-PPARγ decreased significantly in model group compared with that in control group (P〈0.01), while PGE2 mRNA increased (P〈0.01). The indexes above were alleviated in the three TCM-treated groups and aminophylline group, while tiaobu feishen therapies were better than aminophylline. Conclusion: The three tiaobu feishen therapies and aminophylline can improve oxidative stress in the lungs of rats with COPD at different degrees, and tiaobu feishen therapies show more benefits than aminophylline, in which Bufei Yishen Granule has better effect than the other two TCM granules.
