乳岩宁方减轻阿霉素对MCF-7荷瘤裸鼠机体损伤实验研究

Study on Ruyanning plus ADM to Induce Apoptosis of MCF-7 Breast Cancer and Improve General State on Tumor-bearing Nude Mice

目的 :观察乳岩宁方减轻化疗药阿霉素(ADM)对乳腺癌裸鼠一般状态的损伤。方法 :MCF-7细胞株接种于BALB/C裸鼠右侧胸壁第二乳垫部皮下,10 d后选取荷瘤成功的28只裸鼠随机分为对照组、化疗组(ADM)、乳岩宁组及结合组(乳岩宁+ADM),每组7只。每天记录裸鼠体重变化情况;运用ZZ-6小鼠自主活动测试仪检测荷瘤裸鼠活动能力;实验结束处死裸鼠取脾脏,称重。结果 :乳岩宁组小鼠体重显著高于化疗组,结合组显著高于化疗组,有统计学差异(P<0.05);乳岩宁组及结合组的自主活动次数明显高于化疗组(P<0.01),脾重化疗组最轻,结合组次之,乳岩宁组最重(P<0.05)。结论 :乳岩宁方具有修复ADM对荷瘤裸鼠一般状态的损伤并改善生存质量的作用。

Objective： The aim of this study was to observe the influence of Ruyanning plus ADM to affect the quality of life. Method： Nude mice were injected MCF-7 cell on the secondary breast pad of the right chest wall subcutaneously. 28 mice were randomized into model group （ MG ）, chemotherapy group （ ADM, AG ）, Ruyanning group （ RG ） and combination group （ Ruyanning＋ADM, CG ） after MCF-7 tumor-bearing successfully. The change of weight was recorded every day. The ability of activity was detected by ZZ-6 Locomotor activity in mice tester. At the end of the experiment, the mice were killed, and the spleen was taken out and weighed. The pathomorphologial apoptosis was observed by TEM. Results： Weights of mice in RG were higher than that of CG and AG （ P〈0.05 ）; automatic activities in RG and CG were higher than tht of AG obviously （ P〈0.05 ）; weight of spleen in AG was the lightest, while in RG was the heaviest. The apoptosis was induced by Ruyanning plus ADM through TEM. Conclusion ： Ruyanning can enhance ADM effect to induce apoptosis, repair the injury caused by ADM and improve the quality of life on ttnnor-hearing nude mice.
