电针联合BM SC_s移植治疗对出血性脑卒中大鼠血清BMP表达的影响 被引量:1

The Effect of Electro-acupuncture Combined with BMSCs Transplantation on the Expression of Myelin Basic Protein in the Serum of Rats Following Intracerebral Hemorrhage

摘要 目的:探讨电针联合骨髓间充质干细胞(BMSCs)移植治疗对出血性脑卒中大鼠血清髓鞘碱性蛋白(MBP)表达的影响。方法:将培养的BMSCs和生理盐水分别移植入脑出血模型大鼠尾壳核内,将NSS评分≥9分的模型大鼠随机分为4个实验组:生理盐水组(NS组);电针组(Ea组);BMSCs移植组(BMSCs组);电针联合BMSCs移植组(Ea BMSCs组),每组25只。另选正常对照组(Normal组)5只,不做任何处理;假手术组(FO组)5只,造模和移植都同步麻醉和进针但不注入任何药物。用MK3全自动酶标仪对血清中MBP进行ELISA检测。结果:脑出血各治疗组MBP第1天表达最高,随即下降第3天达谷底,随后快速上升第7天达到波峰后下降,但都高于FO组和Normal组(P<0.05)。与NS组比较:Ea组第5、7、14天MBP表达低于NS组(P<0.05);BMSCs组MBP表达第14天低于NS组(P<0.05);Ea BMSCs组第5、7、14天MBP表达显著低于NS组(P<0.01)。与Ea BMSCs组比较:Ea组第1、3天MBP表达低于Ea BMSCs组(P<0.05),第14天高于Ea BMSCs组(P<0.05);BMSCs组MBP表达第5、7、14天高于Ea BMSCs组(P<0.05)。结论:电针联合BMSCs移植治疗脑出血能有效保护神经组织减轻脱髓鞘反应,降低MBP的表达。 Objective:To investigate the effect of electro-acupuncture combined with BMSCs transplantation on the expression of Myelin basic protein in the serum of rats following intracerebral hemorrhage Method:Cultured BMSCs and normal saline were injected into caudate nucleus of rats following intracerebral hemorrhage(ICH)separately. The model rats’ NSS scores ≥ 9 points were randomly divided into four experimental groups:physiological saline group(NS group);curative group(Ea group);BMSCs transplantation group(BMSCs group);acusector joint BMSCs transplantation group(Ea BMSCs group),each group of 25 only. Chosen normal control group(Normal group)5 only,did not do any processing;control group(FO)5 only,made mould and transplant synchronous anesthesia,but no drug injection. MBP expression in serum was tested by ELISA. Result:MBP of treatment groups was the highest on 1 d ,and reduced to the lowest level on 3 d,then increased rapidly and reached the peak on 7 d. The content of MBP in all treatment groups were higher than that of the FO group and the normal group(P〈0.05). The content of MBP in the Ea group was lower than that of the NS group on 5,7,14 d(P〈0.05). The content of MBP in the BMSCs group was lower than that of the NS group on 14 d(P〈0.05). The content of MBP in the Ea BMSCs group was lower than that of the NS group on 5,7,14 d(P〈0.01). The content of MBP in the Ea group was lower than that of the Ea BMSCs group on 1,3 d(P〈0.05),and was higher than that of the Ea BMSCs group on 14 d(P〈0.05). The content of MBP in the BMSCs group was higher than that of the Ea BMSCs group on 5,7,14 d(P〈0.05). Conclusion:BMSCs transplantation combined electro-acupuncture treatment of cerebral hemorrhage can effectively alleviate demyelination reaction to protect the nerve tissue ,and reduce the expression of MBP.
