摘要 目的探讨溶血及宫颈黏液对人乳头状瘤病DNA检测结果的影响。方法对含不同浓度血红蛋白(Hb)或宫颈黏液的宫颈脱落细胞样本,用Real-time PCR法进行HPV DNA检测。结果 Hb浓度为5、10 g/L的溶血样本组与未溶血样本相比HPV DNA Ct值结果差异无统计学意义(P>0.05);但冻融解黏组、NaOH解黏组与黏液未处理组相比HPV DNA Ct值结果减小,差异有统计学意义(P<0.05)。结论 HPV DNA扩增检测结果未受样本溶血影响,而样本中含黏液对HPV DNA检测结果有影响,但经冻融或NaOH解黏处理后可降低复检率。 Objective To evaluate the impact of hemolysis and cervical mucus for HPV DNA detection. Methods By using Realtime PCR, HPV DNA were detected from cervical epithelium samples containing different concentrations of hemoglobin or cervical mucus. Results The resμLts of HPV DNA Ct value were no obvious different between non-hemolytic epithelium samples and treated with different concentrations of Hb (5, 10 g / L)(P〉0.05); The resuLts of Ct value from cervical mucus samples treated by the process of freeziug and thawing or NaOH were lower than untreated, the difference was statistically sinificant (P〈0.05). Conclusion The resuLts of HPV DNA were not affected by hemolysis, but influenced by cervical mucus, and the rate of re-examination could be reduced by treatment with the process of freezing and thawing or NaOH.
