摘要
本文报道血液中碳氧血红蛋白基于吸光度差值的双波长分光光度检测方法。检血用含有 Na2 S2 O4 的溶液稀释得到 Hb CO和 Hb的双组分溶液 ,Hb有相同吸光度的双波长处的吸光度差值只反映 Hb CO的浓度 ,Hb CO有相同吸光度的双波长处的吸光度差值只反映 Hb的浓度 ,测定了这些吸光度差值可以利用给定的公式计算出检血的 Hb CO%。进行比较实验 ,当检血是由 Hb CO和 Hb溶液混合制备时 ,本方法测定值与理论值严格一致 ,当检血是中毒案件检血时 ,本方法测定值与可靠方法的测定值严格一致。
This paper described the spectrophotometric determination of carboxyhemoglobin in blood on the basis of double wavelength spectrophotometry of absorbance difference. Blood was diluted with a Na 2S 2O 4 solutinon to provide a solution of two components of HbCO and Hb. Absorbance difference at the two wavelengths at which Hb has same absorbance reflects only the HbCO component and absorbance difference at the other two wavelengths at which HbCO has same absorbance reflects only the Hb component.After measurement of this absorbance differences,the percentage of HbCO in blood sample is calculated from the absorbance differences using a giving formula. The results of the present method are in close agreement with theoretical values while the samples were mixed with HbCO and Hb. The results are in close agreement with that of reliable method while the samples are case specimen.
