氨基胍对CTGF在大鼠纤维化肺成纤维细胞中表达的影响及其作用机制探讨

摘要 目的探讨氨基胍对结缔组织生长因子(CTGF)在大鼠纤维化肺成纤维细胞中表达的影响及其作用机制。方法采用细胞培养、免疫组化、流式细胞术等方法,检测氨基胍对CTGF在大鼠纤维化肺成纤维细胞中表达的影响。结果 TGF-β可诱导肺成纤维细胞分化为肌成纤维细胞,并使CTGF的表达增强(P<0.05),氨基胍可以抑制TGF-β的诱导作用(P<0.05),单独应用氨基胍对CTGF在大鼠纤维化肺成纤维细胞中的表达无影响。结论氨基胍可能是通过与TGF-β发生作用,从而抑制CTGF在大鼠纤维化肺成纤维细胞中的表达。 Objective It is to approach the influence and action mechanism of aminoguanidine on the expression of connective tissue growth factor (CTGF) in rats with fibrosis lung fibroblast. Methods By cell culture, immunocytochemistry, flow cytometry and so on, the influence of aminoguanidine on the expression of CTGF in rats with fibrosis lung fibroblast were detected. Results The transforming growth factor betal (TGF - β) could induced lung fibroblast differentiate to muscle fibro- blast, and made the expression of CTGF to strengthen ( P 〈 0.05 ) , aminoguanidine could restrained the induce action of TGF - β ( P 〈 0.05) , but single use of aminoguanidine had no influence for the expression of CTGF in rats with fibrosis lung fibroblast. Conclusion It may be the aminoguanidine through the interaction with TGF - β, to the aim of restraining the expression of CTGF in rats with fibrosis lung fibroblast.
