摘要
目的了解合肥地区HIV/AIDS未治疗人群基因亚型和原发性耐药情况。方法应用巢式RT-PCR扩增HIV-1 pol区聚合酶基因,经DNA测序后进行系统进化树分析和耐药性分析。结果收集79例合肥地区HIV/AIDS未治疗者,经RT-PCR扩增并获得有效基因亚型序列65例,基因分型以CRF01-AE重组亚型最多39例(60.0%);其次依次是CRF07-BC 21例(32.3%)、B亚型4例(6.2%)和CRF08-BC1例(1.5%)。1例蛋白酶抑制剂(PIs)主要耐药基因突变,2例反转录酶抑制剂(RTIs)耐药基因突变。总耐药突变率为4.6%(3/65)。结论当前合肥市主要的HIV-1流行亚型是CRF01-AE和CRF07-BC,HIV原发性耐药的发生率在国内处于较低水平。
Objective To understand the genetic subtypes and drug resistance associated mutations of HIV-1 stains currently circulating among antiretroviral treatment-naive patients in Hefei.Methods Seventy-nine HIV-1-infected treatment-naive individuals living in Hefei were sampled and their plasma was collected.HIV-1 pol gene were amplified by nest RT-PCR,sequenced and phylogenetically analyzed.Drug-resistance associated mutations in protease and reverse transcriptase regions were analyzed with Standford University HIV/AIDS Drug Resistance Database.Results CRF01-AE predominated in Hefei with 60.0%,followed by CRF07-BC(32.3%),subtype B(6.2%),CRF08-BC(1.5%).The mutation conferring primary and secondary resistance to protease inhibitors(PIs) were detected in 1(1.5%) of 65 treatment-naive individuals; whereas the mutations conferring resistance to reverse transcriptase inhibitors(RTIs) were identified in 2(3.1%) of 65 treatment-naive individuals.The whole rate of drug resistance mutation was about 4.6%(3/65).Conclusions Major genetic subtypes of HIV-1 in Hefei at present were CRF01-AE and CRF07-BC.The prevalence of genotypic drug resistance was low among treatment-naive HIV-1 infected patient in Hefei.
