新型布尼亚病毒安徽分离株全基因组测序分析 被引量:6

Sequence analysis for full length genomes of Severe fever with Thrombocytopenia Syndrome Bunyavirus in AnHui Province

摘要 目的对安徽省两株新型布尼亚病毒进行全基因组序列测定,分析其全基因组序列的基因特征与变异特点。方法应用22对测序引物对病毒基因组进行RT-PCR分段扩增,PCR产物直接进行序列测定,ClustalW、DNASTAR、MEGA4.1等软件分析基因组序列。结果两株病毒全基因组编码情况和已报道新型布尼亚病毒相同,未见插入和缺失;构建系统发生树可见AHZ毒株与河南省毒株进化距离最近,而AHL毒株在一支较为独立的分支上;M片段膜蛋白前体编码区氨基酸变异率最高。结论安徽省新型布尼亚病毒毒株基因组相对稳定,可能存在与河南省毒株相互传播的现象。 Objective To get the full length genomes of severe fever with thrombocytopenia syndrome bunyavirus ( SFTSV ) isolated in Anhui province in 2011, and to analyze the genomes and variation characteristics. Methods The vi- ral genomes were amplified by RT - PCR method with 22 pairs of primers, and the products of the amplification DNA were sequenced, and the sequences were analyzed by the software of ClustalW, DNASTAR, MEGA4.1 et al. Results The vi- res genome coding of two Anhui strains was same to the strains reported, with no insertion and deletion. The AHZ strain was more closed related to Phlebovirus HN6, but the AHZ strain was in a relatively independent branch. The mutation rate of the M segment was highest, which encoded the membrane protein precursor. Conclusion The genomes of SFTSV in An- hui province were relatively stable, and the mutual spread with Henan might exist.
