摘要
以乌贼喉软骨为原料,碱酶法提取,提取液经TCA和超滤处理,得到硫酸软骨素;经DEDA-52纤维素离子交换柱纯化和sephdex G-25分离,得到CS-1和CS-2两个样品。经紫外光谱扫描,CS-1和CS-2在260 nm和280 nm的核酸和蛋白质吸收峰处均没有吸收,说明样品中不存在核酸和蛋白质。CS-1和CS-2经红外光谱扫描,其图谱基本一致,且在856 cm-1和819 cm-1附近都有振动峰,初步判定是E型硫酸软骨素。样品CS-1和CS-2经凝胶层析法确定分子质量为39 800 u和19 500 u。
Using squid cartilage as raw material,the paper researches the methods on extracting,identifying and purifying Chondroitin sulfate,moreover,analyses of cs quality and properties.A method of dilute alkali-enzyme TCA precipitation to extract chondroition sulfate from the squid was studied.DEAE-52 ion exchange chromatography,two kinds of CS marked CS-1 and CS-2 was separated.Both CS-1 and CS-2 had no protein and nucleic acid left and their purity were shown to be on a single elution peak by UV spectrum.The FTIR spectras of CS-1 and CS-2 from cuttlefish extracts exhibited peaks at both 856 cm-1and 819 cm-1indicating that these CS consisted of both chondroitin-4-sulfate and chondroitin-6-sulfate,therefore preliminary judge squid chondroitin sulfate is E type chondroitin sulfate.The average molecular weight of CS-1 and CS-2 were 39 800 u and 19 500 u.
