摘要
以菊芋幼茎、块茎为试验材料,比较不同激素浓度配比对外植体培养的影响研究.结果表明:幼茎是试验的最佳外植体,愈伤组织诱导快,且有腋芽发生;不定芽分化适宜培养基是MS+BA 2.0mg/L+NAA 0.01mg/L,不定芽分化率100%;在增殖培养基中不定芽增殖系数达3.25,切分后的茎段分化腋芽或不定芽的增殖系数平均为9.6;不定芽在MS+NAA 0.01mg/L培养基上生根率为93%,试管苗经炼苗后移栽成活率达到91.5%.
The effects of MS mediums supplemented with various concentrations of hormones on the proliferation and rooting of Jerusalem artichoke were studied.Its tuber tissue and young stem were used as explants for screening high frequency of adventitious shoot regeneration.The results indicated that the induction rate of adventitious shoot rate from young stem was 100% on MS medium with BA 2mg/L and NAA 0.1mg/L.The propagation coefficient of adventitious shoots was 3.25on the multiplicative culture medium,a 9.6-fold increase per generation.93%of the regenerated adventitious shoots rooted on the rooting medium(MS+ NAA 0.01mg/L).Survival rate of the regenerated plantlets,which were hardened off,was as high as 91.5% when they were transplanted on to the field.
