力生长因子、E肽重组腺病毒的构建及其在神经细胞中的表达

摘要 力生长因子(mechano-growth factor,MGF)是新近发现的一种诱导性表达的生长因子,由IGF-1基因选择性剪接产生,力生长因子E肽(MGF-24E)是MGF区别于其它IGF-1剪接体的特异性C端序列,其在预防组织修复等方面有重要作用.采用Ad-Easy法成功构建MGF,MGF-24E重组腺病毒,重组腺病毒分别感染神经胶质细胞N9和神经元细胞SH-SY5Y,MTT法检测其对两种细胞增殖的影响.结果表明,经腺病毒介导可实现MGF和MGF-24E在神经细胞中的高表达,与对照组相比,MGF和MGF-24E在时间和浓度上对促进N9和SH-SY5Y细胞的增殖均有显著性效应,且二者效果相当.由此认为,MGF和MGF-24E具有促进神经损伤修复的同等作用力,为力生长因子用于基因治疗提供更为切实的理论基础. Mechano-growth factor(MGF)is a newly discovered inducible growth factor produced from IGF-1gene by alternative splicing.MGF-24E is a specific C-terminal sequence of MGF different from other IGF-1 spliceosomes.They both play an important role in the prevention of myocardial injury and nerve repair.In this study,we constructed a recombinant adenovirus containing MGF and MGF-24E gene.Then we used the recombinant adenovirus to infect N9 and SH-SY5Y cells and applied MTT to test their effects on cell proliferation.The result showed that compared with the control group,both MGF and MGF-24E had remarkable concentration and time effects on the proliferation N9and SH SY5Y cells.It is concluded that MGF and MGF-24E have the potential function to promote the repair of nerve injury,which would provide more practical and theoretical basis in gene therapy of mechano-growth factor.
