九龙藤黄酮对大鼠心肌缺血再灌注损伤的保护作用 被引量:7

Protective effect of Jiulongteng flavonoids on myocardial ischemia reperfusion injury in rats

摘要 目的:探讨九龙藤黄酮(Bauhinia championii Benth Flavonoids,BCBF)对心肌缺血再灌注损伤(myocardial ischemia reperfusion injury,MIRI)的保护作用。方法:48只SD大鼠,随机分为假手术、模型、维拉帕米、BCBF低、中、高剂量组(60,120,240 mg·kg-1)共6组(n=8)。结扎冠状动脉左前降支30 min,再灌注60 min建立MIRI模型。观察造模前后大鼠心率(HR),左心室舒张末期压(LVEDP),左心室内压最大上升/下降速率(±dp/dtmax)等血流动力学指标的变化;测定肌酸激酶同工酶(CK-MB)、超氧化物歧化酶(SOD)、丙二醛(MDA)、Na+-K+-ATPase、Ca2+-Mg2+-ATPase含量及心肌梗死范围。结果:与模型组相比,BCBF能剂量依赖性地降低HR、LVEDP及MDA、CK-MB的含量,减少心肌梗死面积;升高±dp/dtmax、SOD,增加Na+-K+-ATPase、Ca2+-Mg2+-ATPase活性(P<0.05或P<0.01)。结论:BCBF对MIRI大鼠具有显著的保护作用。 OBJECTIVE To investigate the protective effect of Bauhinia charnpionii Benth flavonoids (BCBF) on myocardial ischemia reperfusion injury (MIRI) in SD rats. METHODS 48 rats were randomly divided into six groups (n = 8) : sham opera- tion, MIRI, verapamil and low, middle, high dose of BCBF (60, 120, 240 mg·kg^-1 ). MIRI model was established by ligating the left anterior descending coronary artery for 30 min and then reperfusing for 60 min. Hemodynanic indexes such as heart rate ( HR), left ventricular end-diastolic pressrure (LVEDP) and the maximum rate of increase/decrease of left ventricular pressure (dp/dtmax) were observed. Meanwhile, CK-MB, SOD, MDA, Na^+ -K^+ - ATPase, Ca^2+ - Mg^2+ -ATPase and the areas of myocardial infarction were detected. RESULTS BCBF decreased HR, + dp/dt MDA and CK-MB, reduced myocardial ischemia sizes, while increased LVEDP, SOD and the activity of Na^+- K^+ -ATPase and Ca^2+ - Mg^2+ -ATPase (P〈0. 05 or P〈0. 01 vs model control) with dose dependent manner. CONCLUSION BCBF has significantly protective effects on myocardial ischemia . reperfusion injury.
