摘要
目的:探讨胺碘酮对人肝细胞株HL-7702生长的影响。方法:以人正常肝细胞系HL-7702为模型,用MTS法测定胺碘酮对该细胞生长的影响;倒置显微镜法观察给药后细胞形态的改变;流式细胞仪测定细胞周期的变化。结果:MTS实验表明胺碘酮能抑制HL-7702细胞增殖,并表现出浓度、时间依赖性关系。在10-100μmol·L^-1浓度范围内,胺碘酮能剂量依赖性地抑制人肝细胞株HL-7702的生长,其中25,50,100μmol·L^-1胺碘酮抑制细胞生长作用具有统计学意义(P〈0.01)。镜下观察经胺碘酮处理的HL-7702呈现出细胞形态学改变,细胞明显皱缩,之间相互连接消失,可能形成少量类似凋亡小体的物质。细胞周期测定显示处G1期的细胞比对照增多,S期细胞减少。结论:胺碘酮可显著抑制人肝细胞株HL-7702的生长,其抑制细胞的生长可能是使肝细胞阻滞于G1期,使进入DNA复制S期的细胞减少,从而影响细胞的增殖。
OBJECTIVE To investigate the effects of amiodarone(AM) on growth of hepatic cell line HL-7702. METHODS The inhibitory effect of AM on growth of hepatic cell line HL-7702 was measured by MTS assay. The morphology of HL-77(12 was observed by inverted microscope. The cell cycle was analyzed by flow cytometry. RESULTS The results of MTS assay re veaied that AM could inhibit the proliferation of HL-7702 in a dose- and time-dependent manner. HL-7702 cells were incubated in RPMI 1640 medium,which were incubated for 24h and 48h after adding AM in different concentrations(25,50,100 μmol·L^-1 ). The A value of the cells decreased significantly(compared with the control group, all P〈0. 01 ). Results under the invert ed microscope showed that the cell morphology changed after injected with AM; the cells were shrinked significantly, the connections between cells were disappeared,and the number of cell was decreased. The results of flow cytometry showed that HL- 77{}2 cells at G1 phase were increased obviously but S phase decreased, compared with the control group. CONCLUSION AM can delay HD7702 at G1 phase and significantly inhibit the proliferation of HL-7702 cell.
