通莲汤对胃癌MGC803细胞增殖及细胞周期的影响 被引量:2

Effection of Tonglian Decoction on Cell Proliferation and Cell Cycle of Cell Line MGC803 of Gastric Carcinoma

摘要 目的研究通莲汤(TD)对胃癌MGC803细胞形态和生长周期的调节作用;探讨其抑制胃癌细胞增殖的机制。方法将MGC803细胞于37℃,5%饱和湿度的CO2培养箱孵育24 h,分别加入倍比梯度的TD、TD化裁方(THC)和不同浓度的消癌平注射液(XAP),48h后以MTT比色法检测细胞增殖变化、光镜观察细胞形态、流式细胞技术(FCM)测定细胞周期。结果 TD的IC50值为60.2μg·mL-1;与空白对照组(C)相比,该方可显著抑制细胞增殖,影响细胞形态;流式细胞术分析表明,TD组细胞在G2期的比率为(69.56±2.06)%。结论通莲汤通过抑制细胞进入M分裂期从而干预胃癌MGC803细胞生长,与消癌平对该细胞系的调节周期作用相类似。 Objective To investigate effect of Tonglian decoction (TD) on proliferation of gastric carcinoma cell line MGC803 through determination on cell morphology and cell cycle; to discuss inhibition mechanism of the prescription on gastric cancer. Methods Human gastric carcinoma cell line MGC803 cultured in vitro treated with TD and its modified formula (THC), and Xi- ao - ai - ping Injection (XAP) as the positive controlled drug. Cell proliferation is assayed by MTT, cells morphology is observed with microscope and cell cycle is measured with Flow Cytometer (FCM). Results Dose for 50 percent cells inhibition rates ( ICsD ) of TD is 60.2 p^g ~ mL- i. Compared with blank controlled group, TD can inhibit ceils morphology and proliferation. In TD group, rate of cells in phase G2 of cell eyele gets to (69.56 + 2.06) %. Conclusion TD ean be used to regulate MGC803 eareinoma eells proliferation by inhibiting cells entering M phase, having similar mechanism with XAP, whieh is widely used to anti -gastrie eaneer in clinic.
