摘要
从鸭肝中提取脂肪酸合成酶,研究了脂肪酸合成酶的提取条件及温度、pH、常见金属离子对其活性的影响。结果表明匀浆方式为运行10 s停20 s、缓冲液温度为4℃、酶保存温度为-40℃时获取的酶液活性最高;酶活力分别在温度为5℃,pH为7.0时表现出最高;加入金属离子钙、镁、钾后,酶的平均活力升高,可能有促进酶促进作用;加入金属离子钠、铜、锌后,随浓度增加活力呈现下降趋势,可能具有酶抑制作用;加入铁离子的时候随浓度增加出现波浪变化,对酶活力没有影响作用。
Fatty acid synthetase (FAS) was extracted from duck liver to study the effect of fatty acid synthetase and common metal ions, temperature and pH on its activity. Results show that the slurry approach for nanning 10 s and stopped 20 s, buffer, temperature of 4 °C, enzyme preservation temperature of-40 °C for top liquid enzyme activity. Enzymatic activity in the temperature of 5 °C shows the highest when pH is 7.0. After adding metal ions, calcium, magnesium, potassium, the average enzyme activity increases, enzyme average activity may be promoted. After adding metal ions, sodium, copper, zinc, enzyme average activity presents downlrend with increasing concentration of energy as there may be enzyme inhibition. Wave change appears when add increasing concentration iron ion. which shows it has no effect on enzyme activitv.
