脑胶质瘤中miR-143对MACC1表达的调控作用 被引量:3

摘要 目的探讨脑胶质瘤中miR-143对MACC1基因表达的调控作用。方法实时定量PCR检测脑胶质瘤及癌旁组织中miR-143与MACC1基因的表达。将miR-143的前体pre-miR-143和抑制物anti-miR-143分别转染脑胶质瘤U87细胞,Western blot检测MACC1蛋白的表达。荧光素酶报告基因分析验证miR-143与MACC1基因3′UTR的结合。结果与癌旁组织相比,miR-143与MACC1的表达在脑胶质瘤组织中分别呈现显著的下调和上调,且二者的表达呈显著负相关。转染后pre-miR-143和anti-miR-143能够分别下调和上调脑胶质瘤U87细胞中MACC1蛋白的表达。荧光素酶报告基因分析证实miR-143能够与MACC1基因3′UTR结合。结论 miR-143与MACC1基因的表达改变与脑胶质瘤相关,miR-143能够靶向负性调节MACC1基因的表达。 Objective To study the regulatory effect of miR-143 on MACC1 gene in brain glioma. Methods Real-time PCR was used to detect the expression of miR-143 and MACC1 gene in brain glioma and pefi-cancerous tissues. The pre-miR-143 and anti-miR-143 were transfected to U87 cell respectively, and Western blot was used to detect the expression of MACC1 protein. The lueiferase reporter gene analysis was applied to study whether miR- 143 could bind to 3' UTR of MACC1 gene. Results The expressions of miR- 143 in brain ghoma were much lower than those in pefi-cancerous tissues, and the expressions of MACC1 gene in brain glioma were much higher than those in pefi-cancerous tissues. There was a nega- tive relationship between the expression of miR- 143 and MACC1 gene. After transfection, pre-miR- 143 could significantly down regulate the expres- sion of MACC1 protein significantly, and anti-miR- 143 showed the opposite effect. The lucfferase reporter gene analysis confirmed that miR- 143 could bind to the 3' UTR of MACC1 gene. Conclusion The expression changes of miR- 143 and MACC1 gene were related to brain glioma. MiR- 143 could targetedly negatively regulate MA CC1 gene expression.
