甲状腺乳头状癌组织中CXCR7 mRNA和蛋白表达的生物学意义 被引量:3

摘要 目的研究甲状腺乳头状癌组织中趋化因子受体CXCR7mRNA和蛋白的表达,探讨其表达在甲状腺乳头状癌发生、发展中作用。方法采用实时荧光定量PCR和Westernblot检测79例甲状腺乳头状癌和癌旁组织标本中CXCR7mRNA和蛋白的表达,分析CXCR7表达与临床病理特征之间的相关性。结果甲状腺乳头状癌组织中CXCR7mRNA和蛋白的表达水平分别为1.61±0.80和1.1014±0.6938,明显高于癌旁组织的0.74±0.45和0.0927±0.0961(均P〈0.05)。CXCR7mRNA和蛋白的表达与甲状腺乳头状癌淋巴结转移相关,淋巴结转移组中CXCR7mRNA和蛋白表达水平分别为1.87±0.72和1.4443±0.6710,明显高于非淋巴结转移组的1.30±0.79和0.6917±0.4639(均P〈0.05);CXCR7蛋白表达与TNM分期相关,Ⅲ-Ⅳ期中CXCR7蛋白表达为1.5341±0.6732,明显高于Ⅰ~Ⅱ期的0.9546±0.6421(P〈0.05)。结论CXCR7高表达与甲状腺乳头状癌的淋巴结转移、分期相关,参与了甲状腺乳头状癌的发生、发展。 Objective To investigate the mRNA and protein expression of chemokine receptor CXCR7 in papillary thyroid carcinoma and exploreits biological fimction. Methods Theexpression of CXCR7mRNA and protein in 79cases of papillary thyroid carcinoma and paracancerous tisues was detected by real-time quantitative PCR and Western blot. The association between CXCR7 expression and clinicopathological characteristics in papillary thyroid carcinoma was analyzed. Results The expression of CXCR7 mRNA and protein ( 1.61±0.80, 1.101 4-0.693 8 ) was significantly higher in papillary thyroid carcinoma than in paracancerous tissues (0.74±0.45,0.092 7±0.096 1 ) (P 〈 0.05 ). The expression of CXCR7 mRNAand protein was correlated with lymph node metastasis. The expression level of CXCR7 mRNA and protein in the group with lymph node metastasis ( 1.87±0.72, 1.444 3±0.671 0) was higher than in the group without lymph node metastasis ( 1.30±0.79,0.691 7±0.463 9) (P 〈 0.05 ). The level of CXCR7 protein was relevant to TNM stages. The level of CXCR7 protein in stage III-IV (1.534 1±0.673 2) was higher than in stage I - II(0.954 6± 0.642 1 ) (P 〈 0.05). Conclusion The high expression of CXCR7 was related with lymph node metastasis and TNM stages. CXCR7 may be in- volved in the tumorigenesis and progression of papillary thyroid carcinoma.
