培土生金法对脾虚哮喘大鼠模型水通道蛋白5表达的影响 被引量:12

The Effect of “Reinforcing Earth to Strengthen Metal” on Aquaporin 5 Expression in Rats with Spleen-deficiency in Bronchial Asthma

摘要 目的观察培土生金法对脾虚哮喘大鼠模型肺组织水通道蛋白5(AQP5)表达的影响,探讨脾虚对哮喘气道黏液高分泌形成的影响及培土生金法对其的调控作用。方法 50只健康雄性Wistar大鼠,随机分为5组:对照组、脾虚哮喘组、哮喘组、脾虚哮喘治疗组、哮喘治疗组,每组10只。采用RT-PCR测定大鼠肺组织AQP5 mRNA表达水平,采用免疫组化测定肺组织AQP5蛋白表达水平。结果脾虚哮喘组、哮喘组与对照组比较,脾虚哮喘组与哮喘组比较,大鼠肺组织AQP5 mRNA和蛋白表达水平均显著降低(P<0.01)。脾虚哮喘治疗组与脾虚哮喘组比较,哮喘治疗组与哮喘组比较,肺组织AQP5 mRNA和蛋白表达水平均显著升高(P<0.01)。结论培土生金法能够升高脾虚哮喘和哮喘时肺组织AQP5 mRNA和蛋白表达水平。 Objective To observe the effect of "reinforcing earth to strengthen metal" on aquaporin 5 ( AQP5 ) expression in rats with spleen - deficiency in bronchial asthma, and analyze the relationship of spleen-deficiency with airway mucus hypersecretion formation and the effective mech- anism of the "reinforcing earth to strengthen metal" method. Methods 50 healthy male Wistar rats were randomly divided into 5 groups : control group, spleen-deficiency asthma group, asthma group, spleen-deficiency asthma treatment group, and asthma treatment group (n =10). Expression of A QP5 mRNA in the lung was detected by reverse transcription polymerase chain reaction (RT-PCR), and AQP5 protein expression was detected by immunohistochemistry. Results The expression of A QP5 mRNA and protein in the lung tissue of rats was significantly decreased in spleen defi- ciency asthma group comparing with the asthma group (P 〈 0.01 ). The expression level ofAQP5 mRNA and protein in lung tissue was significantly increased in spleen deficiency asthma treatment group and asthma treatment group comparing with spleen deficiency asthma group and asthma group, respectively (P 〈 0.01 ). Conclusion The expression level of A QP5 mRNA and protein in spleen deficiency asthma or asthma lung tissue can be significantly increased by the treatment of "reinforcing earth to strengthen metal".
