复方中药对免疫抑制小鼠免疫促进作用的研究 被引量:6

摘要 目的从细胞及分子水平,探求中药复方对免疫功能增强的作用及效能,在扶正固本、活血益气的中药中遴选出有广阔应用前景的促进免疫的中药复方。方法 50只昆明小鼠随机分成正常对照组、模型对照组与高剂量中药组、中剂量中药组、低剂量中药组。用环磷酰胺抑制小鼠免疫功能制作免疫抑制模型,观察用药后正常对照组、模型对照组与高剂量中药组、中剂量中药组、低剂量中药组血清IgG、补体C3,脾淋巴细胞表面抗原标志物CD3+、CD4+、CD8+、CD4+/CD8+、CD19+、CD49b+,脾淋巴细胞细胞因子IL-2和IFN-γ及体重和脾指数。结果与模型对照组相比,高剂量中药组、中剂量中药组、低剂量中药组体重和脾指数、CD3+、CD4+、CD8+、CD4+/CD8+、CD19+差异无统计学意义(P>0.05),低剂量中药组血清IgG降低[(177.19±45.66)g/L],CD49b+、补体C3及脾淋巴细胞细胞因子IL-2和IFN-γ升高[(32.43±6.31)%、(294.0±61.2)g/L、(38.44±7.82)%、(22.11±6.29)%],差异均有统计学意义(均P<0.05),中剂量中药组脾淋巴细胞细胞因子IL-2和IFN-γ升高[(35.40±4.59)%、(22.26±8.84)%],差异均有统计学意义(均P<0.05)。正常对照组与模型对照组比较,差异有统计学意义(P<0.05)。结论尽管还未能证实复方中药对特异性细胞免疫和体液免疫有促进作用,但在一定剂量范围内对非特异性细胞免疫和体液免疫有显著促进作用。 Objective To select the new broad application prospect of the Chinese medicine compound for promoting immunity by studying the effect of traditional Chinese medicine compound from cell and molecular levels in Fuzheng Guben and Huoxue Yiqi of traditional Chinese medicine.Methods Fifty Kunming mice were randomly divided into five groups:the normal control group,the model control group,the high dose Chinese medicine group,the middle dose Chinese medicine group and the low dose Chinese medicine group.The model of immunosuppression was established with cyclophosphamide inhibiting the immune function of the mice.Serum IgG,complement C3,splenic lymphocyte surface antigen markers of CD3+,CD4+,CD8±,CD4+/CD8+,CD19+,CD49b+,lymphocyte cell factor IL-2 and IFN-γ,weight of body and spleen index of the mice in the normal control group,the model control group,the high dose Chinese medicine group,the middle dose Chinese medicine group and the low dose Chinese medicine group were observed.Results Compared with the model control group,weight and spleen index,CD3+,CD4+,CD8+,CD4+/CD8+,CD19+ of the mice in the high dose Chinese medicine group,the middle dose Chinese medicine group,the low dose Chinese medicine group had no significant difference,decreasing serum IgG of (177.19±45.66) g/L and increasing CD49b+ of (32.43±6.31)%,complement C3 of (294.0±61.2) g/L,splenic lymphocyte cytokines IL-2 of (38.44±7.82)%and splenic lymphocyte cytokines IFN-γ of (22.11±6.29)%of the mice in the low dose of Chinese medicine group had no significant difference (all P < 0.05),of spleen lymphocytes cytokines IL-2 of (35.40±4.59) % and IFN-γof (22.26±8.84)% of the mice in the middle dose Chinese medicine group increased and there were significant difference (P < 0.05) between the normal control group and the model control group.Conclusion It is still not confimed that the compound Chinese medicine has a promoting effect on specific cellular immunity and humoral immunity,however,compound Chinese medicine dramatically promotes the non-specific cellular immunity and the non-specific humoral immunity in a certain dose range.
