摘要
目的:运用Plackett-Burman 实验设计联用星点设计(central composite design,CCD)效应面法,考察各影响因素显著性,筛选长叶胡颓子黄酮提取的最佳工艺.方法:运用Plackett-Burman 实验设计筛选主要影响因素,采用星点设计-效应面法优选长叶胡颓子黄酮提取工艺.乙醇浓度、料液比、回流温度作为自变量,黄酮含量作为因变量,通过对二者二次响应曲面的回归拟合,利用三维曲面图直观分析长叶胡颓子黄酮提取最佳工艺,并进行预测分析.结果:黄酮的最佳提取工艺为:乙醇浓度80%~85%,料液比为1∶15~1∶20,回流温度60~65℃,回流2次,30min/次.结论:利用Plackett-Burman 实验设计联用星点设计(CCD)确定了长叶胡颓子黄酮的提取工艺,该方法简便,精度更高、重现性好、预测性强.
Objective.To optimize the process of extracting effective constituents from Elaeagnus bockii Diels by Plackett-Burman combined with CCD response surface methodology. Methods : In Plackett-Burman combined with CCD response surface design, Independent variables were concentration of ethanol, Liquid ratio, reflux temperature. Dependent variables were contents of total fla vones. Results. The Optimization extraction of total flavones was 80%-85 % ethanol, 15-20 fold of solvent and 2 times for refluxing extraction at the 60℃ -65℃ ,30 minutes each time. Conclusions: The optimum process is simple and more convenient for extracting the total flavones from Elaeagnus bockii Diels with higher precision and predictability.
