摘要
目的 研究浙东海岛地区动脉粥样硬化性脑梗死患者的血浆脂联素水平、相关基因SNP+45 T/G多态性及两者之间的关系.方法 183例急性脑梗死患者(脑梗死组),男97例,女86例;年龄37 ~ 81岁,平均(62±8)岁.200例体检正常者(对照组),男110例,女90例;年龄40 ~ 76岁,平均(60±7)岁.所有患者于人院24 h内、对照组于体检当日,空腹采静脉血3 ml,1500 r/min离心5 min(离心半径r=10 cm),尔后置于-30℃冰箱待测.用ELISA法测定血浆脂联素水平,限制性片段长度多态性聚合酶链反应(polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)方法检测脂联素SNP+ 45 T/G多态性.结果 脑梗死组杂合子型(TG型)+纯合子突变型(GG型)频率(44.3%)、G等位基因频率(25.4%)均高于对照组(33.0%,18.3%),而血浆脂联素水平[(3.01±0.26)μ/L]低于对照组[(5.86 ±0.37) μg/L].Logistic回归分析发现,脂联素基因SNP +45TG+ GG型与脑梗死的发病正相关,血浆脂联素水平与脑梗死的发病负相关.结论 脂联素基因SNP+ 45 TG+ GG型可能是浙东海岛地区动脉粥样硬化性脑梗死的易感基因型,可能是通过影响血浆脂联素表达引发脑梗死.
Objective To investigate the levels of plasma adiponectin in patients with atherosclerotic cerebral infarction and related gene SNP + 45 polymorphism and the correlation between them in island area of East Zhejiang.Methods The levels of plasma adiponectin were measured with ELISA,and related gene SNP +45 polyrnorphism was monitored with polymerase chain reaction-restriction fragment length polymorphism (PCRRFLP).Results The frequencies of TG + GG genotype and G allele in the cerebral infarction (CI) group were all significantly higher than those in the control group,while the levels of plasma adiponectin in the CI group were lower than those in control group.Logistic regression analysis revealed that the adiponectin gene SNP + 45 TG + GG was positively correlated with that of CI patients,and the levels of plasma adiponectin were negatively associated with CI patients.Conclusions Adiponectin gene TG + GG genotype was possibly a susceptible genotype associated with those patients with atherosclerotic cerebral infarction in island area of East Zhejiang.The occurrence of CI might be resulted from its effect on the expression of plasma adiponectin.
