黄芪对压力超负荷大鼠肾脏UrotensinⅡ及胶原表达的影响

Effect of Astragalus on Expression of Urotensin Ⅱ and Collagen in Renal Tissues of Rats with Heart Failure

摘要 目的:观察黄芪对腹主动脉缩窄致慢性压力超负荷大鼠肾组织中UrotensinⅡ及胶原表达的影响。方法:雄性SD大鼠40只,随机选取10只大鼠为假手术组(Sham组),其余30只制作腹主动脉缩窄模型,随机分为模型组(Model组),黄芪组(Atragalus组)和厄贝沙坦组(Irbesartan组),每组10只。各组大鼠在术后24 h开始给药,连续给药12周后,记录大鼠体重,并检测血清尿素氮(BUN)、肌酐(Scr)及24 h尿蛋白定量;处死大鼠,留取肾组织行HE及Masson染色观察肾间质病理改变,采用免疫组化学法观察UrotensinⅡ、胶原Ⅰ、Ⅲ在肾组织中表达的变化。结果:(1)与假手术组相比,12周末模型组大鼠24 h尿蛋白定量、血清BUN、Scr水平明显增高,差异有统计学意义(P<0.01);黄芪组和厄贝沙坦组的上述指标与模型组比较,则有明显降低,差异有统计学意义(P<0.05)。(2)与模型组相比,黄芪组和厄贝沙坦组肾小管空泡变性、坏死明显减少,结构基本完整,间质胶原沉积减少。(3)与假手术组比较,模型组肾组织UⅡ、胶原Ⅰ、Ⅲ表达增加;与模型组比较,黄芪组和厄贝沙坦组肾组织UⅡ、胶原Ⅰ、Ⅲ表达明显降低。结论:黄芪可能通过降低UⅡ及胶原Ⅰ、Ⅲ在肾间质的过度表达,延缓慢性压力超负荷大鼠肾间质纤维化的进程,进而发挥肾脏保护作用。 Objective:To observe the effect of Astragalus shrink narrow induced chronic stress expression of Urotensin II and collagen in renal tissue of overload in rats on abdominal aorta. Methods:Forty male Sprague -Dawley(SD) rats weighing 200 ~ 220 g were used. Randomly selected 10 rats in sham operation group ( group Sham), the remaining 30 making coarctation of abdomi- nal aorta model, were randomly divided into model group (group Model), Astragalus group (group Atragalus) and the Maher Bbe Chatain group ( Irbesartan group), 10 rats in each group. The rats in 24 h after operation administration, continuous administration for 12 weeks, records of body weight, and serum urea nitrogen (BUN), ereatinine (Scr), and 24 h urinary protein; rats were killed, taken by HE and Masson in renal tissue staining of renal interstitial pathological changes, the immunohistochemical method was used to observe the expression of Urotensin II , collagen I , III in renal tissues. Results: ( 1 ) As compared with Sham group, Model group rats at the end of the 12 week, 24 h urinary protein, serum BUN, Ser levels were significantly higher(P 〈0.01 ) ; the index in Model group compared with Astrugalus group and Irbesartan group, there was decreased ( P 〈 0.05 ). (2) Compared with Model group, in Astragalus group and Irbesartan group, renal tubular degeneration, necrosis was significantly reduced, the basic structure of integrity, interstitial collagen deposition reduction. (3) Compared with group Sham, in renal tissue of Model group, U II , collagen II , III expression increased; compared with Model group, significantly lower U II , collagen I , III expression in renal tissue of Astragalus group and Irbesartan group. Condusion:Astragalus membranaceus may through the decreasing over expression of U II and the collagen I , III in renal interstitial, delaying chronic pressure overload rat renal tubulointerstitial fibrosis, and played a role in renal protection.
